补中益气汤对A549／DDP与A549荷瘤BALB／c小鼠移植瘤生长的影响

摘要

Objective To observe the effect of Buzhong Yiqi decoction on the growth of transplantation tumor of BALB /c mice with lung adenocarcinoma cisplatin-resistant cells (A549 /DDP) and cisplatin-sensitive cells (A549).Methods The lung tumor-bearing model was established by inoculation of A549 /DDP cells and A549 cells into BALB /c nude mice subcutaneously.The mice were treated by intrap-eritoneal injection of cisplatin for 25 d, twice per day.Simultaneously, the animals in traditional Chinese medicine groups also were put into low, medium and high dose of Buzhong Yiqi decoction for 25 d, twice per day.At 0, 5, 10, 15, 20, 25 d,the long and short diameter of transplantation tumor were measured , then the volume was calculated and the growth curve of tumor was drew .At last, all the mice were exe-cuted.The weight of transplantation tumor was weighed , then the tumor inhibitory rate and tumor inhibitory increasing rate were calculated . Results For transplantation tumor with A549 /DDP cells, Buzhong Yiqi decoction reduced the volume of transplantation tumor after 10 d of administration, and lost the weight of transplantation tumor , increased the tumor inhibitory rate and tumor inhibitory increasing rate after 25 d of administration.For transplantation tumor with A549 cells, Buzhong Yiqi decoction reduced the volume of transplantation tumor after 5 d of administration, and lost the weight of transplantation tumor , increased the tumor inhibitory rate and tumor inhibitory increasing rate after 25 d of administration.Conclusions Buzhong Yiqi decoction could inhibit the growth of transplantation tumor of BALB /c mice.It might play a role in a tumor suppressor in the cisplatin-sensitive and cisplatin-resistant lung tumor tissues.%目的观察补中益气汤对 A549／DDP 与 A549荷瘤 BALB ／c 小鼠移植瘤生长的影响。方法BALB ／c 小鼠右腋窝皮下接种 A549／DDP 或 A549细胞株，成瘤后连续腹腔注射顺铂25 d，2次／d，中药治疗组同时连续灌胃低、中、高剂量补中益气汤25 d，2次／d。给药后0、5、10、15、20、25 d，测量移植瘤的长径与短径，计算移植瘤的体积并绘制肿瘤的生长曲线。实验结束后处死小鼠，剥离瘤组织并称重，计算药物抑瘤率及抑瘤作用提高率。结果对于 A549／DDP 移植瘤，给药10 d 后，补中益气汤开始缩小移植瘤体积，随着给药时间的延长和给药剂量的增加，移植瘤体积逐渐缩小；给药25 d 后，随着补中益气汤剂量的增加，移植瘤重量随之减轻，抑瘤率随之增加，抑瘤作用提高率也随之显著增加。对于 A549移植瘤，给药5 d 后，补中益气汤即开始缩小移植瘤体积，随着给药时间的延长和给药剂量的增加，移植瘤体积逐渐缩小；给药25 d 后，随着补中益气汤剂量的增加，移植瘤重量随之减轻，抑瘤率随之增加，抑瘤作用提高率也随之显著增加。结论补中益气汤能抑制 BALB ／c 肺腺癌荷瘤小鼠肿瘤的生长，在顺铂耐药与顺铂敏感的肺癌组织中均可发挥抑瘤作用。
