目的 探讨丹皮酚对大鼠心肌缺血再灌注损伤保护中肿瘤坏死因子(TNF)-α及Toll样受体(TLR)4表达的影响.方法 清洁级成年雄性SD大鼠36只随机分为假手术组、 缺血再灌注组、丹皮酚低剂量组和丹皮酚高剂量组每组9只;通过结扎左冠脉前降支30 min再灌注2 h建立大鼠心肌缺血再灌注模型.通过TTC染色法检测大鼠梗死心肌体积;ELISA法检测血液中TNF-α的含量;通过免疫组化法检测缺血坏死心肌组织TLR4的表达.结果 与缺血再灌注组相比,丹皮酚低剂量组和丹皮酚高剂量组心肌梗死体积减小,TNF-α表达减少;心肌组织TLR4水平明显降低;其中丹皮酚高剂量组更明显.结论 丹皮酚能够明显降低大鼠心肌缺血再灌注损伤,这种作用可能与减轻缺血再灌注中大鼠心肌TNF-α及TLR4表达有关.%Objective To investigate the effect of paeonol ( PA) on the expression of TNF-αand TLR4 in myocardium ischemia (MI)-reperfusion injury (RI) in rats.Methods A total of 36 male SD rats were randomly divided into sham , ischemia reperfusion (IR), low-dose PA and high-dose PA groups , 9 rats in each group .And the rat MI/RI model was established by coronary artery ligation for 30 min followed by 120 min reperfusion .The myocardium infarction was measured by TTC staining .Blood TNF-αwas detected by ELISA .The ex-pression of TLR4 in ischemic myocardium was detected via immuno-histochemistry.Results Compared with IR group , myocardial infarction and the expression of TNF-αand TLR4 were reduced in low-and high-dose PA groups , the effects of high-dose PA were more obvious .Con-clusions PA could significantly reduce myocardial ischemia-reperfusion injury in rats , which may be related to reduce the expression of TNF-αand the expression of TLR 4 in myocardium of ischemia-reperfusion rats .
