草果知母汤对学习记忆功能的影响及与抗癫痫作用的关系

         

摘要

目的 探讨草果知母汤对学习记忆功能的影响及与抗癫痫作用的关系.方法 选择昆明种小鼠90只和P77-PMC 大鼠,即听源性发作(AGS)易感大鼠90只.小鼠分为正常A组、草果知母汤A组和苯巴比妥A组,大鼠分为癫痫模型组、草果知母汤B组和苯巴比妥B组,每组30只,Wistar大鼠30只为正常B组.采用跳台法,按学习记忆的3个过程:获得、巩固、重现分为3大部分观察,测定草果知母汤对正常小鼠和癫痫大鼠学习记忆功能的影响.另选AGS大鼠20只,分为无惊厥组12只和惊厥组8只,测定抗癫痫作用.结果 与正常A组和苯巴比妥A组比较,草果知母汤A组明显提高记忆成绩(P<0.05,P<0.01).与苯巴比妥B组比较,草果知母汤B组明显延长小鼠记忆获得和重现过程的触电潜伏期、提高记忆成绩(P<0.01).与癫痫模型组比较,草果知母汤B组对癫痫大鼠的获得及重现障碍有明显的翻转作用,能明显延长大鼠记忆获得和重现过程的触电潜伏期(P<0.01),提高学习和记忆成绩(P<0.05).无惊厥组与惊厥组大鼠学习记忆各项指标差异无统计学意义(P>0.05).结论 草果知母汤有一定的益智作用,但与其抗癫痫作用无相关关系,为其本身所具有.%Objective To study the effect of Fructus Tsaoko and Rhizoma Anemarrhenae Decoction (FTRAD) on learning and memory and its relation with antiepileptic effect .Methods Ninety P77 PMC rats susceptible to audiogenic seizure (AGS) and 30 Wistar rats included in this study were divided into normal group B , epilepsy model group , FTRAD group B ,and phenobarbital group B (30 in each group ) according to the 3 processes of learning and memory (acquisition ,consolidation and retrieval) .The effect of FTRAD on learning and memory of rats was detected .Another 20 rats susceptible to AGS were divided into non-convulsion group (n=12) and convulsion group (n=8) .Results The latent period of electric shock for memory acquisition and retrieval was sig -nificantly longer and the learning and memory score was significantly higher in FTRAD group B than in phenobarbital group B (P<0 .01) .The effect of FTRAD on reverse of memory acquisition and retrieval was significantly better in FTRAD group B than in epilepsy model group (P< 0. 01), indicating that FTRAD can significantly prolong the latent period of electric shock for memory acquisition and retrieval of rats and improve their learning and memory scores (P< 0. 05) .However ,no significant difference was found in learning and memory between the non-convulsion group and convulsion group (P>0 .05) .Conclusion FTRAD is beneficial for improving intelligence of rats ,which is not related its anti-epileptic effect.
