nHAC/CSH复合犬外周血基质细胞修复种植体周围骨缺损的组织学观察

     

摘要

Objective:The aim of the study was to assess the new bone formation around immediate dental implants using nHAC/CSH loaded with dog blood-acquired mesenchymal progenitor cells (dBMPC) in a canine model. Methods: Dog blood-acquired mesenchymal progenitor cells (dBMPCs) were isolated from peripheral blood of healthy adult dogs. Four healthy mongrel dogs were used in this study. The alveolar bone defects around immediate implants of dogs were created. Each defect was randomly assigned to one of the following three groups:(1) no materials (controls); (2) injectable bone cement: nHAC/CSH; or (3) the injectable tissue engineered bone (ITB) group: nHAC/CSH- dBMPC; Methylene blue staining was used to examine the bone formation after 12 weeks. Results: The nHAC/CSH- dBMPC group showed significantly more bone-implant contact than either nHAC/CSH or control groups in the areas with peri-implant defects12 weeks after implantation. Conclusion: The results indicate that the ITB composed of nHAC/CSH and dBMPC may represent a useful strategy for the clinical reconstruction of bone defects around immediate implantation. However, further investigation is needed involving the use of human BMPC as well as possible use of seeding cells.%目的:评价可注射纳米羟基磷灰石/胶原/硫酸钙(nHAC/CSH)植骨材料复合犬外周血基质细胞修复种植体周围骨缺损的骨形成情况。方法:犬外周血基质细胞(dBMPC)从健康成年犬的外周血中分离得到并行成骨诱导。选用4条成年杂种犬,拔除前磨牙后制备骨缺损模型,缺损处分三组进行处理(1)空白组;(2)单纯材料组(nHAC/CSH)和(3)可注射骨(nHAC/CSH-dBMPC)。12周后处死动物,制备硬组织磨片,行亚甲基蓝染色并计算骨-种植体结合率(BIC)。结果: nHAC/CSH-dBMPC组BIC明显高于其余两组,具有统计学意义。结论: nHAC/CSH和dBMPC组成的可注射组织工程骨为即刻种植体周围骨缺损的临床修复另辟蹊径。但对于人类BMPC的利用还需进行进一步研究。
