N受体对四磨汤诱发的胃窦平滑肌收缩的影响

             

摘要

目的 研究烟碱(N)受体对四磨汤诱发的SD大鼠离体胃窦平滑肌条收缩活动的影响.方法 离体胃窦平滑肌条置于盛有Krebs液的组织浴槽中以记录其等长收缩活动,观察不同剂量四磨汤(1 μL、5μL、25 μL、50 μL、100 μL、150 μL、200 μ,L)对大鼠胃窦平滑肌条收缩活动的影响,并观察N受体阻断剂六烃季胺( 10-4mol/L)对四磨汤引起的胃窦平滑肌收缩活动的影响.结果 四磨汤引起大鼠胃窦纵行和环行平滑肌条收缩活动增加呈剂量依赖性.六烃季胺对于四磨汤引起的胃窦纵、环形平滑肌条兴奋作用均具有部分抑制效应,对环行肌条的作用明显高于对纵行肌的影响.结论 四磨汤对大鼠胃窦平滑肌的收缩活动具有明显的兴奋作用,但部分作用与N受体有关,胃窭环行肌更为依赖N受体的作用.%Objective To investigate the effect of Simo Decoction on contractile activity of isolated antral longitudinal and circular strips of SD rats and its underlying mechanism through N receptor. Methods The contractions of antral longitudinal and circular strips from SD rats were observed in organ bath under baseline or administration of hexametho-nium chloride (10-4 mol/L) respectively, and consecutive concentrations of Simo Decoction (1 μL, 5 μL, 25 μL, 50 μL, 100 μL, 150 μL, 200μL) were put into the bath to observe the strip responses. Results Simo Decoction dose-dependently increased the contractions of longitudinal and circular strips in antrum. Significant differences were observed among different concentrations. After treated with N-receptor inhibitor hexamethonium chloride, the increased contractile activity caused by Simo Decoction in longitudinal and circular strips could be partly inhibited at each concentration, and the inhibitory effect on circular strips was more significant than that on longitudinal strips. Conclusion Simo Decoction can dose-dependently promote the contraction of antral longitudinal and circular muscles, which partly decided by activation of N-receptor, and effect on circular strips is more dependently on N-receptor.
