反义HCCR-2转染对肝癌细胞HepG2形态的影响

         

摘要

目的 研究HCCR-2反义基因转染对肝癌HepG2细胞株形态学的影响.方法 用脂质体法将HCCR-2反义真核表达载体导入HepG2细胞,从光镜、HE染色、荧光显微镜、电镜水平观察转染前后细胞形态学的改变.结果 与亲本细胞相比,光镜及HE染色下转染HCCR-2反义基因的HepG2细胞变得狭长,胞体增大,胞浆丰富,分裂相减少,核浆比下降,异型性减少;荧光显微镜下反义基因转染细胞发出绿色荧光,透射电镜下反义基因转染细胞出现凋亡.结论 HCCR-2反义基因可以促进HepG2细胞的分化.%Objective To investigate the effect of HCCR-2 antisense gene transfection on the morphology of HepG2 cells. Methods HCCR-2 antisense-nucleic acids were transfected into HepG2 cell by LipofectamineTM2000. Morphological changes of the transfected cells were observed by light micriscope, HE staining, fluorescence microscope and transmission electron microscope. Results By light microscope and HE staining, cells transfected with HCCR-2 antisense gene were narrow-longer with abundant cytoplasm and nuclear division decreased, the ratio of nuclei to cytoplasm decreased compared with their parental cells. Green fluorescene of the stable transfected HepG2 cell lines could be observed by inverted fluorescence microscope. Apoptosis could be observed by transmission electron microscope. Conclusion HCCR-2 antisense-nucleic acids could promote differentiation of HepG2 cells.
