免疫抑制剂联合神经生长因子促周围神经损伤修复研究

摘要

Objective To investigate the united effect of immunosuppressant and nerve growth factor on peripheral nerve injury. Method Established peripheral nerve crush injury model, and divided into four groups, saline group (inject saline by vein), NGF group (local added nerve growth factor), immunosuppressant group (inject FK506 by tail vein), NGF + FK506 group (local added nerve growth factor and injected FK506 by tail vein). Then, observed the effect by Catwalk gait analysis system, electrophysiological test and nerve tissue pathology. Result The results of Catwalk showed that the recovery of nerve function in NGF group and FK506 group were better than saline group, but worse than NGF+FK506 group. Similarly, electrophysiological test and nerve tissue pathology also showed that the effect of NGF + FK506 group in peripheral nerve injury repair was better than other groups. Conclusion Immunosuppressant, combined with nerve growth factor can promote the repair of nerve injury, there is a synergistic effect between the two.%目的探讨免疫抑制剂联合神经生长因子(NGF)促进周围神经损伤修复的可行性。方法建立周围神经crush损伤模型,并按照分组要求分为尾静脉注射生理盐水(saline组)、局部添加NGF (NGF组)、尾静脉注射免疫抑制剂(FK506组)、局部添加NGF联合尾静脉注射免疫抑制剂(NGF+FK506组)。然后,借助于Catwalk步态分析系统、电生理检测及神经组织病理学等方法评估免疫抑制剂联合NGF促周围神经损伤修复的效果。结果 Catwalk步态分析系统测试结果提示NGF组和FK506组的恢复效果优于saline组,与NGF+FK506组相比较差;电生理测试证明了NGF+FK506组的神经肌肉电生理恢复情况优于其他3组;神经病理学同样也证明NGF+FK506组的促神经损伤修复效果优于其他3组。结论免疫抑制剂联合NGF可以促进神经损伤的修复,二者存在协同作用。
