系统性红斑狼疮患者外周血IL-17、IFN-γ的表达及其与疾病活动性的关系

ObjectiveTo study the expression of IL-17 and IFN-γ in peripheral blood of patients with systemic lupus erythematosus (SLE), and explore their correlations with disease activity.MethodAccording to SLE disease activity index (SLEDAI), 78 patients with SLE were classiifed into active group (SLEDAI>9, 31 patients) and inactive group (SLEDAI≤9, 47 patients). ELISA was used to analyze the expression of IL-17 and IFN-γ in peripheral blood of patients with SLE and 50 healthy controls.ResultThe expression of IL-17 and IFN-γ in peripheral blood of patients with SLE were significantly higher than control group (P<0.05). The expression of IL-17 and IFN-γ in active group were significantly higher than those of inactive group (P<0.05). The levels of plasma IL-17 and IFN-γ showed a positive correlation with SLEDAI (P<0.05). Compared with that without renal damage, SLE patients with renal damage had higher levels of IL-17 and IFN-γ (P<0.05). After treatment for 4 weeks, the expression of IL-17 and IFN-γ in active group were signiifcantly decreased than those before and after treatment for 2 weeks (P<0.05).ConclusionThe expression of IL-17 and IFN-γ in peripheral blood of patients with SLE is over-expressive, and closely related with disease activity. IL-17 is expected to become new targets for the treatment of SLE.%目的：探讨系统性红斑狼疮（systemic lupus erythematosus，SLE）患者外周血白细胞介素-17（IL-17）、干扰素-γ（IFN-γ）的表达及其与疾病活动性的相关性。方法将78例SLE患者根据SLE疾病活动性指数（SLEDAI）评分分为稳定期（SLEDAI≤9分）47例，活动期（SLEDAI＞9分）31例。采用酶联免疫吸附测定（ELISA）检测SLE患者及健康体检者（对照组，50例）血浆中IL-17、IFN-γ的水平。结果 SLE患者外周血IL-17、IFN-γ表达水平较对照组显著升高（P＜0.05），且活动期患者IL-17、IFN-γ表达水平显著高于稳定期（P＜0.05）；SLE患者IL-17、IFN-γ表达与SLEDAI均呈线性正相关（P＜0.05）；与无肾损伤者相比，有肾损伤的SLE患者IL-17、IFN-γ表达水平明显升高（P＜0.05）；治疗后4周，活动期SLE患者外周血IL-17、IFN-γ表达水平较治疗前、治疗后2周均显著下降（P＜0.05）。结论 SLE患者外周血IL-17、IFN-γ呈过度表达状态，且与疾病活动性密切相关， IL-17有望成为SLE治疗的新靶点。
