人脐带间充质干细胞移植对大鼠薄型子宫内膜的治疗作用

摘要

Objective To investigate the effect of human umbilical cord-derived mesenchymal stem cells (hUC-MSCs) whether can repair the thin endometrium of rats, and the possibility of repairing damaged endometrium with mesenchymal stem cells. Method Established rat thin endometrium model. 16 healthy sexually mature female SD rats were randomly divided into 4 groups, rats with hUC-MSCs transplantation via veins in the ovulation induction period;rats with PBS transplantation in the ovulation induction period;rats with hUC-MSCs transplantation via veins in the non-ovulation induction period and rats with PBS transplantation in the non-ovulation induction period. Removal of the uterine endometrium after three estrous cycles, compared expression of keratin, vimentin and integrinβ3 of each group. Result 12 days after hUC-MSCs transplantation, the endometrial thickness of every group [non-ovulation PBS group (679.12±186.20) pixels,non-ovulation transplantation group (969.39±367.44) pixels, ovulation PBS group (820.35±153.99) pixels, ovulation transplantation group (841.58±242.32) pixels] showed no significant difference (P>0.05). After the hUC-MSCs was transplanted for 12 days, the average optical density values of the keratin (0.01551, 0.00340, 0.00229, 0.00160), vimentin (0.02348, 0.04758, 0.02052, 0.02552) and integrinβ3 (0.00283, 0.00531, 0.00966, 0.00154) among ovulation transplantation group, non-ovulation transplantation group, ovulation PBS group and non-ovulation PBS group were significantly different (P < 0.05). Conclusion hUC-MSCs may improve endometrial receptivity by induction function and immunoregulation. The two transplantation periods both have different degree of efficacy in improving the thin endometrium.%目的探讨人脐带间充质干细胞(human umbilical cord-derived mesenchymal stem cells,hUC-MSCs)移植能否修复薄型子宫内膜模型大鼠的子宫,研究间充质干细胞修复受损内膜的可能性。方法建立SD大鼠子宫内膜模型。将健康性成熟未交配雌性SD大鼠16只随机分为促排卵周期移植组、促排卵周期磷酸盐缓冲液(PBS)组、非促排卵周期移植组、非促排卵周期PBS组,每组各4只,于3个动情周期后处死大鼠,取其子宫内膜标本,比较各组大鼠角蛋白、波形蛋白以及整合素β3的表达情况。结果 hUC-MSCs移植后12天各组大鼠子宫内膜厚度分别为:非促排卵周期PBS组(679.12±186.20)像素、非促排卵周期移植组(969.39±367.44)像素、促排卵周期PBS组(820.35±153.99)像素、促排卵周期移植组(841.58±242.32)像素,组间比较均无显著差异(P>0.05)。hUC-MSCs移植后12天,促排卵周期移植组、非促排卵周期移植组、促排卵周期PBS组、非促排卵周期PBS组大鼠角蛋白平均光密度值分别为0.01551、0.00340、0.00229、0.00160,波形蛋白平均光密度值分别为0.02348、0.04758、0.02052、0.02552,整合素β3平均光密度值分别为0.00283、0.00531、0.00966、0.00154,组间比较差异均具有显著性(P<0.05)。结论 hUC-MSCs可能通过诱导作用和免疫调节特性改善子宫内膜的容受性。两个移植时期对改善薄型子宫内膜均有不同程度的疗效。
