tRNA鸟嘌呤糖基转移酶在肺腺癌组织中的表达及临床意义

摘要

目的 探究tRNA鸟嘌呤糖基转移酶(tRNA-guanine transglycosylase,TGT)在肺腺癌组织中的表达及临床意义.方法 蛋白质免疫印迹法分析TGT在肺腺癌细胞系A549细胞和肺正常细胞系人支气管上皮细胞(16HBE细胞)中的表达情况;免疫电镜观察TGT在A549细胞和16HBE细胞中的分布情况;免疫组织化学法分析TGT在肺腺癌组织和正常组织中的表达情况.结果 蛋白质免疫印迹法分析结果显示:TGT在A549细胞和16HBE细胞中均有表达,且TGT在A549细胞中高表达.免疫电镜观察可见TGT在A549细胞和16HBE细胞的线粒体、粗面内质网、高尔基体、细胞质及细胞核中均有分布,QTRT1(QTRT1为鸟嘌呤插入酶,代表TGT)在A549细胞线粒体中的表达高于其在16HBE细胞线粒体中的表达.免疫组织化学分析结果显示:TGT在20例肺腺癌患者组织中的阳性表达率显著高于其在正常肺组织中的阳性表达率(P<0.05).结论 TGT在肺腺癌中具有良好的特异性,其对肺腺癌的发生具有一定的预测价值.%Objective To investigate the expression and value of tRNA-guanine transglycosylase (TGT) in lung adenocarcinoma. Method Western blotting was used to analyze the expression of TGT in A549 cells and 16HBE cells; immune electron microscope was used to analyze the distribution of TGT in A549 cells and 16HBE cells; immunohistochemistry was used to analyze the expression of TGT in lung adenocarcinoma and normal tissues. Result Western blotting results showed that, TGT was expressed in A549 cells and 16HEB cells, the expression of TGT in A549 cells was higher than that in 16HBE cells; immune electron microscope results showed that TGT distributed in mitochondrion, rough endoplasmic reticulum, Golgi apparatus, cytoplasm and nucleus in A549 cells and 16HBE cells, the expression of QTRT1 in mitochondrion of A549 cells was higher than that in 16HBE cells. Immunohistochemistry results showed that the positive rate of TGT in 20 patients with lung adenocarcinoma was significantly higher than that in normal lung tissues (P < 0.05). Conclusion TGT has a good specificity in the diagnosis of lung adenocarcinoma, it has a certain early warning value for the development of lung adenocarcinoma.
