抗-HBs定量方法的改进

摘要

Objective Through detecting the standard preparation with series of concentration to indirectly calculate the anti-HBs concentration of the serum samples,a suitable anti-HBs quantitative method for our laboratory was found after comparing the two kinds of methods.Methods Detecting the anti-HBs standard preparation with series of concentration by RIA method,standard curvilinear equatiions were obtained by the means of fitting the detected result and the corresponding concentration by log-log model and exponential curve model respectively.Then the fitting efficiency of two curves was compared.By calculating the concentrations of the reference using two standard curvilinear equations,we can compare the accuracy of two quantitative methods.Result The error mean square of the exponential curve model is low as 1.2971 and the determinate coefficient is close to 1 with the vglue of 0.9904.The average concentrations(n=6)of the detected reference calculated by two eurvilinear equation with the actual concentration of 30.0mlU/mL aIe(32.28±1.06)and(31.91±1.06)mIU/mL respectively.The concentration calculated by exponential curve model is 04 6.37% higher than the actual concentration.Conclusion Fitting by exponential curve model is more practical to estimate the actual concentration of the serum samples those will be detected.It can be used as an optimal quantitative method to detect anti-HBs concentratinn.%目的 通过对系列已知浓度标准品的检测来间接进行血样抗-HBs的定量,比较两种计算方法的准确程度,从而选出适合本实验室的抗-HBs定量方法.方法 用RIA法检测抗-HBs系列已知浓度标准品,将测定结果与对应的浓度分别用log-log模型和指数曲线模型拟合得到标准曲线方程,比较两种曲线拟合的效果.再将对标定浓度质控品的测定值分别代入上述两种模型的曲线方程计算出质控品的浓度,比较两种计算方法定量结果的准确程度.结果 指数曲线模型的误差均方较小,为1.2971;而且确定系数R~2为0.9904,接近1.通过两种曲线方程计算的标定浓度(30.0 mIU/ml)、质控品平均浓度(n=6)分别为(32.28±1.06)和(31.91±1.06)mIU/ml,用指数曲线模型计算的浓度与标定浓度相比仅偏高6.37%.结论 用指数曲线模型拟合能够更好地估算待检样品的实际浓度,可作为本实验室抗-HBs的一种改进定量方法.
