鸡血藤醇提物及其活性成分儿茶素对辐射损伤小鼠脾脏JAK/STAT通路的影响

         

摘要

Objective: To study the effect of ethanol extracts from Spatholobus suberectus and its active component catechin on JAK/STAT pathway in spleen of radiation injured mice. Methods: ICR mice were randomly divided into normal group, model group, positive control group (amifostine, 43.6 mg·kg-1, iv, 30 min before irradiation), SSD group (10, 20, 40 g·kg-1) and catechin group (50, 100 mg·kg-1). Except for normal group, all mice were exposed to 6.0 Gy 60Coγ irradiation with a dose rate of 117.29 cGy·min-1and a duration of 290 s. The mice were administered the appropriate drugs once a day after irradiation for 28 consecutive days. Blood samples were collected from the tail end and the number of peripheral blood cells was counted with a microcell counter before irradiation and on the 28thday. On the 28thday after irradiation, total proteins in spleen cells of mice were detected to study the expression of related protein using Western-blotting and total RNA in spleen cells of mice to show the expression of related gene with RT-PCR method. Results: Spatholobus suberectus and catechin both show activating-hematopoiesis effect. Compared with normal group, related protein in JAK/STAT signal pathway (P < 0.05) and the expression of mRNA (P < 0.01) were significantly up-regulated, the expression of mRNA of Bax and Caspase-3 markedly reduced, and the expression of Bcl-2 distinctly promoted in both Spatholobus suberectus and its active component catechin groups. Conclusion:Spatholobus suberectus and its active component, which have a certain ability to repair the spleen cells of mice, could activate JAK/STAT signal channel and inhibit apoptosis of spleen cells in mice with radiation injury.%目的: 研究鸡血藤醇提物及其活性成分儿茶素对辐射损伤小鼠脾脏的影响.方法:将ICR小鼠随机分为正常组,模型组,阳性对照组(氨磷汀,43.6 mg·kg-1,iv),鸡血藤醇提取物低、中、高剂量组(生药10、20、40 g·kg-1),儿茶素低、高剂量组(50、100 mg·kg-1),除正常组外,各组均接受6.0 Gy剂量60Coγ射线一次性全身照射(剂量率117.29 cGy·min-1,时间290 s),连续灌胃给药28 d,每日1次.于照射后第28天,检测小鼠外周血WBC、RBC、PLT计数,提取小鼠脾脏细胞中的总蛋白, Western blotting法测定相关蛋白表达情况,提取小鼠脾脏中总RNA,采用RT-PCR法测定相关基因表达情况.结果:鸡血藤醇提物和儿茶素均可促进辐射小鼠外周血象的恢复,与正常组相比,均可显著升高JAK/STAT信号通路中相关蛋白表达(P <0.05)和上调mRNA的表达(P < 0.01),均可显著下调Bax和Caspase-3的mRNA的表达(P < 0.01),上调Bcl-2的表达(P < 0.01).结论:鸡血藤醇提物及其活性成分对小鼠脾脏中JAK/STAT信号通路起激活作用,可抑制脾脏细胞的凋亡,对辐射损伤小鼠脾脏具有一定的修复能力.
