血浆和组织 miR-124表达与大肠癌患者预后的相关性分析

         

摘要

目的:分析血浆和组织中miR-124表达水平与大肠癌患者预后的相关性。方法收集大肠癌患者71例,其中无法切除患者49例、完全切除患者22例,收集其血浆和肿瘤组织样本。检测肿瘤无法切除患者血浆和组织中miR-101、miR-124、miR-126、miR-210、miR-217的表达,通过实时定量反转录聚合酶链反应( qRT-PCR)分析miR-124表达和患者及无进展生存期( PFS)的相关性。结果 qRT-PCR检测无法切除大肠癌患者血浆和组织中miRNAs表达,其中高表达的miR-124变化最显著(分别为P <0).05, P <0.01),其他miRNAs无明显变化。血浆中miR-124与患者OS相关:在无法切除大肠癌患者血浆、组织中高表达miR-124与OS呈正相关(血浆:OR=0.451,95% CI 0.189~0.654, P =0.002;组织:OR=0.295,95%CI 0.493~1.374, P =0.037)。 Cox比例风险模型评估:采用Cox比例风险模型评估发现血浆中高表达miR-124与PFS相关(OR=0.147, P =0.022);而组织中高表达miR-124与PFS无相关性(OR=0.281, P =0.057),血浆和组织中低表达miR-124与低死亡风险比无相关性(OR=2.07, P =0.163)。高miR-124表达同PFS无相关性(OR=1.85, P =0.725)。结论血浆中miR-124表达可以作为评价大肠癌预后的生物标记物。%Objective To analyze the correlation between plasma and tissue miR-124 expression and overall survival ( OS) of patients with colorectal cancer.Methods Collected 71 cases of patients with colorectal cancer which cannot be re-moved ( n =49) , completely resected ( n =22) , collected the plasma and tumor tissue samples.Detected tumor unresect-able tissue and plasma of miR-101, miR-124, miR-126, miR-210, miR-217’ s expression, by quantitative real-time reverse transcription polymerase chain reaction ( qRT-PCR) to analyze the correlation between miR-124 expression and the prognosis of the patients.Results qRT-PCR for detection of unresectable expression of miRNAs in plasma and tissue in patients with colorectal cancer, high expression of miR-124’s changes revealed as the most significant ( P <0.05, P <0.01), and other miRNAs did not show significant changes.(1) Correlation between plasma miR-124 and patients’ OS:in unresectable color-ectal cancer patients, plasma and tissue expression of miR-124 was positively correlated with the OS ( OR=0.451, 95% CI 0.189 -0.654, P =0.002;OR=0.295, 95%CI 0.493-13.74, P =0.037).(2) Cox proportional hazards models as-sessment:the Cox proportional hazards model evaluation revealed that the high expression of miR-124 in plasma were correla-ted with PFS ( OR=0.147, P =0.022); and high expression of miR-124 in tissue had no correlation with PFS ( OR=0.281, P =0.057), plasma and tissue low expression of miR-124 had no correlation with low mortality risk ratio (OR=2.07, P =0.163).High expression of miR-124 had no correlation with PFS (OR=1.85, P =0.725).Conclusion It demon-strated that the plasma miR-124 expressions could be used as biological markers for evaluating the prognosis of colorectal cancer.
