目的 研究γ-干扰素(IFN-γ)诱导银屑病皮损角质形成细胞的丝裂原活化蛋白激酶(MAPK)活性,以及MAPK在银屑病发病中的意义.方法 在培养的银屑病患者及正常人角质形成细胞中加入IFN-γ、genistein及二甲基亚砜( DMSO)进行刺激,检测MAPK的活性.结果 银屑病皮损角质形成细胞中MAPK活性较正常人高,且在IFN-γ的刺激下活性均增加,而加入genistein时两组MAPK活性明显下降,两组接受IFN-γ、genistein刺激后,MAPK活性变化的比较差异有统计学意义(P<0.01).结论 IFN-γ对银屑病角质形成细胞MAPK活性有明显促进作用.%Objective To study the effects of interferon (IFN)-γ on mitogen-activated protein kinase (MAPK) activities of cultured psoriatic keratinocytes, and also the significance of MAPK in the pathogenesis of psoriasis. Methods The cultured keratinocytes of psoriatic patients and the normal controls were stimulated by adding IFN-γ, genistein and dimethylsulfoxide (DMSO) in. Then the activity of MAPK was detected. Results The activity of MAPK in psoriasis keratinocytes was higher than that in normal controls group, and the activity of MAPK was increased under the stimulation of IFN--γ. While, the activity of MAPK of both two groups was decreased when adding genistein in. The differences between these two groups were significant (P<0.05). Conclusion IFN-γ could significantly strengthen the MAPK activity of psoriatic keratinocytes.
