创伤性深静脉血栓形成中纤溶相关基因表达变化的实验研究

         

摘要

Objective To study the changes of genes expression related to fibrolysis in rat models of the traumatic deep vein thrombosis( TDVT) by Genechip Rat Genome 4302.0, and to explore the significance of fibrolysis in the TDVT. Methods 30 rats were divided into control( group A) and experiment group. The experiment group were divided into 2 groups according to biological states:thrombosis at the crest - time ( group B) and without thrombosis at the crest - time ( group C) . To incise the femoral vein in corresponding time and adopt Trizol one - step method for total RNA extraction of femoral vein. After applying Genechip Rat genome 4302.0 genechips to detect the RNAs expressions,the genes related to fibrolysis were selected through genechip data analysis. Results Compared with control group, many genes were differential expressions in the process of TDVT, and 65 genes were related to fibrolysis - antifibrolysis system. In which, 42 genes had functional notations, 13 genes of them were promoting fibrolysis and 29 genes of them were antifibrolysis; 23 genes had no exactly functional notation, 8 genes of them could be related to promoting fibrolysis and 15 genes of them could be related to antifibrolysis. The critical genes, such as serpinel, plat, plau, plaur were differential expressions in the process of TDVT. Conclusion In the process of TDVT, the genes related to fibrolysis were intimated with the biological states of thrombi.%目的 应用基因芯片技术研究创伤性深静脉血栓(TDVT)形成过程中纤溶相关基因的表达变化规律,探讨其在TDVT形成中的意义.方法 将30只SD大鼠随机分为正常对照组(A组)和模型组.模型组根据造模后的不同生物学状态再分为两组:高峰期血栓形成组(B组)、高峰期血栓不形成组(C组),在相应时相点无创切取股静脉血管组织,随后抽取各组大鼠总RNA,用Genechip Rat Genome 430 2.0芯片测定股静脉RNA的表达,并分析纤溶基因表达变化情况.结果 在深静脉血栓形成演化过程中,与正常对照组比较,多个基因呈差异表达.共找到与促纤-抗纤相关基因65个,其中已知功能基因42个(促纤13个,抗纤29个);未知基因23个,包括可能与促纤相关基因8个,可能与抗纤相关基因15个.serpine1、plat、plau、plaur等关键基因在该过程中呈差异表达.结论 在TDVT形成中,纤溶相关基因与血栓生物学状态关系密切.
