小鼠重组Periostin对成骨细胞附着和伸展的影响

摘要

目的：观察小鼠重组Periostin对成骨细胞附着和伸展的作用。方法：采用固相结合分析实验和四唑盐比色(MTT)法等细胞生物学技术，观察成骨细胞在Periostin基质上的附着能力和伸展率。结果：成骨细胞在Periostin基质上附着和伸展良好，当浓度在20μg/ml 以上时，作用尤其显著（P＜0.01），与在纤维结合蛋白（Fn）基质上生长相似。结论：小鼠重组Periostin可有效促进成骨细胞的附着和伸展，但其作用的确切分子机制还需深入研究。%AIM: To investigate the effects of mouse recombinant Periostin on adhesion and spread of osteoblast. METHODS: Solid-phase binding assay and MTT method were used to observe the ability of adhesion and spread of osteoblast on Periostin matrix. RESULTS: Adhesion and spread of osteoblast are very well on Periostin at dose of 20μg/ml or more (P<0.01) and the effects of Periostin are similar to FN. CONCLUSION: Periostin can promotes the adhesion and spread of osteoblast, but the mechanism needs further study.
