复发性视神经脊髓炎患者外周血记忆T细胞与IL-7水平的变化

         

摘要

Objective:To observe the changes of memory T cells (CD4+ CD45RA- T cell)and IL-7 level in peripheral blood of patients with relapsing or non-relapsing neuromyelitis optica,and discuss the role of memory T cells in NMO relapsing mechanism.Methods:The proportion of CD4+CD45RA-cells in the relapsing NMO group (n= 15),the non-relapsing NMO group (n= 17 )and the healthy control group (n= 15 )was detected by flow cytometry.Plasma interleukin 7 (IL-7)levels were measured by enzyme-linked immunosorbent assay (ELISA)in all 3 groups.The correlation of CD4+CD45RA- cell ratio with IL-7 level in 32 patients was analyzed.Results:The CD4+CD45RA-cell ratio in the relapsing group (32.86±5.53)%was higher than that in the non-relapsing group (19.98±5.72)%and the healthy control group (19.59± 4.49)%,and the differences were statistically significant (P<0.01);the ratio in the non-relapsing group was higher than that in the control group,but the difference was not statistically significant.The IL-7 level in the relapsing group ([418.73 ± 78.96]pg/mL)was higher than that in the non-relapsing group ([313.33±57.65]pg/mL)and the healthy control group ([303.43±40.48]pg/mL),and the differences were statistically significant (P<0.01);the IL-7 level in the non-relapsing group was higher than that in the healthy control group,but the difference was not statistically significant.The CD4+CD45RA- cell ratio was significantly correlated with IL-7 level in NMO patients (r= 0.817,P= 0.000 ).Conclusions:Memory T cells participate in the relapsing process of neuromyelitis optica (NMO),while IL-7 promotes the cytopoiesis and function of memory T cells.%目的:观察复发与未复发视神经脊髓炎(neuromyelitis optica,NMO)患者外周血记忆T细胞(CD4+CD45RA-T细胞)和IL-7水平的变化,探讨记忆T细胞在NMO复发机制中的作用.方法:利用流式细胞仪检测复发组(15例)、未复发组(17例)NMO患者及健康对照组(15例)CD4+CD45RA-细胞的比例.采用酶联免疫吸附法(ELISA)检测3组血浆白细胞介素7(IL-7)的浓度.分析32例患者的CD4+CD45 RA-细胞比例与IL-7水平的相关性.结果:复发组CD4+CD45 RA-细胞比例(32.86±5.53)%高于未复发组(19.98±5.72)%及健康对照组(19.59±4.49)%,差异均有统计学意义(P<0.01);未复发组CD4+CD45RA-细胞比例高于健康对照组,但差异无统计学意义.复发组IL-7水平[(418.73±78.96)pg/mL]高于未复发组[(313.33±57.65)pg/mL]及健康对照组[(303.43±40.48)pg/mL],差异均有统计学意义(P<0.01);未复发组IL-7水平高于健康对照组,但差异无统计学意义.NMO患者CD4+CD45RA-细胞比例与IL-7的水平显著相关(r=0.817,P=0.000).结论:记忆T细胞参与NMO的复发过程,而IL-7对记忆T细胞的生成和功能有促进作用.
