假丝酵母E-2产生物表面活性剂摇瓶发酵条件的优化

             

摘要

从扬子石化的废水淤泥中筛选到1株能发酵液体石蜡产脂肽类生物表面活性剂的假丝酵母Candida E-2.通过单因子实验和正交试验,得到了最佳发酵培养基组成(g/L):牛肉膏3.0,蔗糖2.0,酵母膏0.25,KH2PO4 12.5,MgSO4 0.3,NaCl 1.5,CaCl,0.05,尿素0.5 5;液体石蜡10%(体积分数).最佳培养条件:初始pH7.0,接种量0.12g/L,装液量为200mL三角瓶30mL,培养时间为5 d.最终产量提高了2.7倍,达1.582g/L.%Candida E-2, a lipopetide biosurfactant producing strain, was isolated from sludge of wastewater treatment factory of Yangzi Petroleum Co. Ltd. The optimized medium composition by the single-factor experiment and orthogonal experiment was obtained (g/L) : beef extract 3. 0,sucrose 2. 0,yeast extract 0. 25, KH2PO4 12. 5, MgSO4 0. 3, NaCl 1.5, CaCl2 0. 05, urea 0. 5, initial pH 7.0, and liquid paraffin 10% ( V/V). The optimized medium condition were inoculum amount 0. 12 g/L, fermentation volume 30 mL/200 mL, fermentation time 5 d. Under such conditions the biosurfactant yield could rise 2. 7 times and reach 1. 582 g/L.
