一株桦褐孔菌伴生菌的鉴定及对肿瘤细胞的抑制作用

         

摘要

为了从药用真菌伴生菌中挖掘具有开发前景的活性菌株,通过显微形态观察结合ITs序列分析,对分离自桦褐孔菌的一株伴生菌进行鉴定,采用细胞四唑盐(MTT)法测定伴生菌和桦褐孔菌对肿瘤细胞的抑制作用.结果表明:桦褐孔菌伴生菌为顶枝孢属的一个种Acremoniumsp.,并可能是一个新种;100 μg/mL的伴生菌和桦褐孔菌提取物对肺癌细胞NCI-H460的抑制率为47.88%和41.32%,对胃腺癌细胞SGc-7901抑制率分别为42.64%和36.26%,对人肝癌细胞HepG2抑制效果差;而稀释10倍的伴生菌发酵液对肺癌细胞NCI-H460和胃腺癌细胞SGc-7901抑制率分别为56.81%和46.63%.%In order to screen active strains from medicinal fungi,we used microscopic morphology and internal transcribed spacer (ITs) sequence analysis to classify the companion fungus separated from Inonotus obliquus,and studied the inhibition of the companion fungus and Inonotus obliquus fermentation on tumor cells by the 3-[4,5-dimethylthiazol-2-yl]-2,5-diphenyl-tetrazolium bromide (MTT) colorimetric assay.The companion fungus is Acremonium sp.,and may be a new species.The extract of the companion fungus and Inonotus obliquus could inhibited 47.88% and 41.32% NC2-H460 cells,42.64% and 36.26% gastric adenocarcinoma cells SGC-7901,respectively.However,the extract of the companion fungus and Inonotus obliquus inhibited hardly human hepatoma cell HepG2.When the broth of the companion fungus was 10 times diluted,it inhibited 56.81% NCI-H460 cells and 46.63% SGc-7901 cells.
