Purpose A pre-column derivatization method by HPLC was established for the determination of free amino acids in Weilening Tablets. Methods The sample was mixed with 2-amino butyric acid as the internal standard, then derivatized with 6-aminoquinolyl-N-hydroxysuccinimidyl carbamate (AQC) and analyzed on a Kromasil C18 column(250 mm × 4.6 mm ,5 μm) ,with ammonium acetate solution( pH 5.10) and acetonitrile solution as mobile phase,using gradient elution with detection at 248 nm. Results The correlation coefficients between the ratios of peak area of amino acid to those of the internal standard and the amino acid concentrations were above 0.999. The average recoveries of amino acids added were 91.5％ -107.0％ and RSD was 0.63％ -3.79％. Conclusion The method could be used for determination of content of free amino acids in Weilening Tablets.%目的:建立柱前衍生高效液相色谱法测定胃乐宁片中各种游离氨基酸的方法.方法:以α-氨基丁酸为内标,以6-氨基喹啉基-N-羟基琥珀酰亚氨基甲酸酯(AQC)为柱前衍生化试荆,采用Kromasil C(18)(250mm×4.6mm,5μm)色谱柱,醋酸铵缓冲液(pH5.10)和57%乙腈水溶液为流动相,梯度洗脱,检测波长为248nm,同时分离测定胃乐宁片中21种游离氨基酸.结果:21种氨基酸在0.01-0.5 mmoL/L浓度范围内有良好的线性关系.r均不小于0.999,平均加样回收率为91.5%-107.O%,RSD为0.63%-3.79%.结论:建立的方法可用于检测胃乐宁片中游离氨基酸含量,测定的结果为胃乐宁片质量研究和评价提供了依据.
