肾虚雄性大鼠红细胞膜组分及跨膜信息传递变化的实验研究

     

摘要

The constitution and function of erythrocyte membrane and the changes of transmembrane information transmission in rats of kidney asthenia were investigated.In this study,a model of male rat of kidney asthenia was established by excision of sexual gland.The activities of protein kinase C (PKC),Ca2+-Mg2+-ATPase,Mg2+-ATPase in erythrocyte membrane and content of membrane phospholipid were observed.The results show that the activities of protein kinase C(PKC)and Ca2+-Mg2+-ATPase in erythrocyte membrane in the model group are significantly lower than those in the control group(P<0.01).The contents of PC and PI in erythrocyte membrane are significantly lower(P<0.01),that of PS is significantly lower (P<0.05) and that of SM is significantly higher(P<0.01).The kidney-tonifying traditional Chinese drugs can increase the activities of protein kinase C(PKC) and Ca2+-Mg2+-ATPase,decrease the content of SM(P<0.01) and increase the content of PC,PI,PS(P<0.01).It is concluded that the change of constitution and function of membrane in male rats of kidney asthenia can have effect on the asymmetry,fluidity and transmembrane information transmission.The kidney-tonifying traditional Chinese drugs can increase the activities of some of enzyme in membrane,correct some of constitution of membrane,such as SM,PC,PI,which can restore the constitution and function of membrane and the transmembrane information transmission,thus have the preventive and therapeutic effect on the diseases of kidney asthenia.%目的:探讨肾虚对细胞膜的结构和功能及其跨膜信息传递的影响以及补肾中药的作用机制。方法:通过手术去势切除性腺造成雄性大鼠肾虚动物模型,施加补肾中药,以红细胞膜蛋白激酶C(PKC)、Ca2+-Mg2+ATP酶、Mg2+ATP酶活性变化和膜磷脂组分含量为观察指标。结果:肾虚模型对照组红细胞膜蛋白激酶C(PKC)、Ca2+-Mg2+ATP酶活性明显低于正常对照组(P<0.01);红细胞膜磷脂组分PC、PI与正常对照组比较,均有非常明显的降低(P<0.01),PS明显降低(P<0.05),而SM明显增高(P<0.01)。补肾中药可使肾虚雄性大鼠的PKC、Ca2+-Mg2+ATP酶活性明显提高(P<0.01);明显降低SM含量,明显增高PC、PI、PS含量(P均<0.01)。结论:肾虚雄性大鼠具有细胞膜结构和功能的改变,可能影响膜的不对称性和流动性及跨膜信息传递。补肾中药具有增强细胞膜的某些酶活性,纠正部分膜磷脂(如SM、PC、PI等)组成成分的作用,使细胞膜的结构和功能、跨膜信息传递趋于正常,对肾虚相关疾病具有防治作用。
