天麻钩藤饮对自发性高血压大鼠血清Ca2+浓度及血管平滑肌细胞钙通道的影响

     

摘要

目的:探讨天麻钩藤饮对自发性高血压大鼠(SHR)的血压及血管平滑肌细胞钙通道电生理特征的影响,阐明天麻钩藤饮对血压干预的作用机制.方法:选用12周龄自发性高血压雄性大鼠45只,随机分为5组.治疗4周后测量收缩压,用全自动图像分析系统进行分析,测定血清游离钙浓度,用全细胞模式膜片钳技术记录分析血管平滑肌细胞L型电压依赖性钙通道的特性.结果:①天麻钩藤饮组用药4周后SHR血压下降最明显(P<0.01);②用药前后天麻钩藤饮组血清游离钙浓度改变没有统计学意义(P>0.05);③天麻钩藤饮组有明显减少血管平滑肌细胞Ica-L内流的作用.结论:①天麻钩藤饮和硝苯地平能够抑制早期的高血压,天麻钩藤饮长期降压效果优于硝苯地平;②天麻钩藤饮有明显的阻滞血管平滑肌细胞L型钙离子通道的作用,这可能是其降压作用机制之一.%Objective: To investigate the effect of Tian ma Gou teng Yin on blood pressure,the electricity physiology feature of L-type calcium channels in the vascular smooth muscle cells in SHR, to explain the mechanism of Tian Ma Gou Teng Yin to blood pressure. Methods; 12-week-old SHRs were assigned randomly into five groups. After 4-week-treatment, systolic blood pressure(SBP) ,and serum free calcium concentration were measured,the electricity physiology feature of L-type calcium channels in the vascular smooth muscle cells was analyzed by patch clamp technique(PCT). Results; ①After 4-week-treatment, SBP was decreased in Tian ma Cou teng Yin group (P <0. 01) ;②No significant difference of Tian ma Gou teng Yin was found in the serum free calcium concentration (P <0. 05) ;③Tian ma Gou teng Yin can degradute the L-type calcium channels current of vascular smooth muscle cells. Conclusions; ①Both Tian ma Gou teng Yin and Nifedipine could repress early phase hypertension while no apparent statistical significance was found between them. The long-term antihypertensive effect of Tian ma Gou teng Yin is superior to Nifedipine;②Tian ma Gou teng Yin can block the L-type calcium channels current of vascular smooth muscle cells, which maybe one of the mechanisms of its antihypertensive function.
