Objective:To investigate The effect and possible mechanism of Puerarin on the cyclin related proteins of oxLDL induced proliferating vascular smooth musle cells. Methods:To establish rabbits' proliferating VSMC model induced by oxLDL, incubate the model with different concentrations of Puerarin for 48 hours,then test the cell cycle, cyclin E and p27 by flowmetry. Results:the amount of G0/G1 stage cells increased in puerarin group while the S stage cells decreased and it has a significant difference between the groups( P < 0. 01 ). The expression of cyclin E decreased while the expression of p27 increased in the puerarin group. Conclusion:puerarin inhibit the proliferation of VSMC, the mechanism may related to cyclin E and p27.%目的:研究葛根素对氧化低密度脂蛋白(oxLDL)诱导增殖的血管平滑肌细胞(vascular smooth muscle cell,VSMC)细胞周期的影响及其相关机制.方法:建立oxLDL诱导的兔VSMC增殖模型,与不同浓度的葛根素共同培养48h后,用流式细胞仪检测VSMC细胞周期变化及细胞周期调控相关蛋白E(cyclin E)和p27的表达.结果:葛根素组G0/G1期细胞数明显增多,S期细胞数明显减少,与LDL组相比差异显著(P<0.01);cyclin E表达量随葛根素剂量增加而减少.p27蛋白表达量则明显增加.结论:葛根素抑制VSMC增殖,其机制与下调cyclin E、上调p27蛋白表达有关.
