脾虚1号方对脾虚型功能性消化不良大鼠肝脏细胞色素C氧化酶Ⅳ的影响

     

摘要

目的:探讨脾虚型功能性消化不良(FD)大鼠肝组织细胞色素C氧化酶Ⅳ(COX Ⅳ)蛋白与基因表达及脾虚1号方干预.方法:70只10d龄雄性SD大鼠按随机数字表法分为正常对照组(10只)、FD模型组(单模型组10只)、脾虚型FD模型组(50只).正常组给予2%蔗糖溶液灌胃,FD模型组和脾虚型FD模型组均给予0.1%蔗糖碘乙酰胺蔗糖溶液灌胃,0.2 mL/只·d,连续6d.脾虚型FD模型组正常饲料喂养至6周龄后叠加改良小平台站立,连续14d;造模结束后按随机数字表法分为双模型组、多潘立酮组、脾虚1号方低、中、高剂量组各10只,药物干预14 d,采用免疫组化、Western-blot和RT-PCR方法检测肝脏组织COXⅣ蛋白与mRNA表达量.结果:与正常组比较,双模型组大鼠肝脏COX Ⅳ mRNA表达量和平均光密度值显著降低;与双模型组比较,脾虚1号方高剂量组大鼠肝脏COX Ⅳ mRNA和蛋白表达量升高.结论:脾虚型FD大鼠存在COX Ⅳ mRNA和蛋白表达量降低现象,脾虚1号方可能是通过上调COX Ⅳ蛋白与mRNA的表达提高脾虚证大鼠能量代谢.%Objective:To investigate the expression of COX Ⅳ protein and mRNA in Liver tissue of Dyspepsia rats with spleen deficiency type and the intervention by Pixu Ⅰ Recipe.Method:70 10-day-old male SD rat pups were randomly divided into normal group(n =10),FD model group (single model,n =10),FD with spleen deficiency model group (n =50).The normal control group were gavaged with 0.2 mL of 2% sucrose,meanwhile,the FD with spleen deficiency model group were gavaged with 0.2 mL of 0.1% IA in 2% sucrose,for 6 days.To postnatal 43 day,the FD with spleen deficiency model group was given modified multiple platform method(MMPM),continued 14h every day,for 14 days.The FD with spleen deficiency model group were averagely randomly divided into double model group,domperidone group,low-dose,medium-dose and high-dose Chinese herbs group,and treated by drug for 14d.To detect the COX Ⅳ protein expression in liver tissues by immunohistochemistry,Western-blot and RT-PCR.Results:Compared with the normal group,the mRNA and protein expression of COX Ⅳ of double model group were significantly decreased;Compared with the double model group,the mRNA and protein expression of COX Ⅳ of high-dose Pixu Ⅰ Recipe group were increased.Conclusion:The FD Rats with Spleen Deficiency had the phenomenon of low expression of COX Ⅳ mRNA and protein,and Pixu Ⅰ Recipe could increase the expression of COX Ⅳ protein and mRNA,and then to promote metabolic energy.
