针刺对端粒酶敲除小鼠大脑海马区、皮质区Trk-B信号调节激酶表达的影响

     

摘要

Objective:To investigate the acupuncture and electric acupuncture intervention that the different acupuncture method in the clinical commonly used,have expression influence on B (Trk-B) signal conditioning tyrosine kinases of the hippocampus region and cortex region in telomerase-deficient (Terc-/-) mice brain.Methods:A total of twenty-four (Terc-/-) mice were randomly assigned to acupuncture group,electric acupuncture group and control group.In the electric acupuncture group,used a pair of hua tuo brand eletroacupunture apparatus (0.3 mm × 0.25 mm) acupuncture at the stomach 36 (ST-36) position,the eletroacupunture apparatus were connected with alternation of frequency and intensity of 0.5 ~ 1.2 mA,2/100 HZ,for 30 min/d for 7d.in the acupuncture group,used a pair of hua tuo brand stainless steel needles (0.3 mm x 0.25 mm)acupuncture at the stomach 36 (ST-36) position,once every five minutes twisting for 30 min/d for 7d.the control group was used in reared under the same conditions without any intervention.Then for Western blot method on the Trk-B expression in the hippocampus region and cortex region.Results:In the hippocampus region,compared with the control group,the electric acupuncture group and the acupuncture group were increased significantly in the Trk-B expression;However,in the cortex region,compared with the electric acupuncture group,the acupuncture group and the control group were decreased significantly in the Trk-B expression.Conclusion:The electric acupuncture intervention has different influence on the expression of Trk-B in the different brain regions,and it can promote the expression of the hippocampus Trk-B;There are different mechanisms in the acupuncture and the electric acupuncture intervention when we acupuncture the same acupoint,it may be related to the expression of Trk -B signal conditioning tyrosine kinases.%目的:探讨临床常用的不同针刺方法,手针、电针干预对端粒酶敲除小鼠大脑海马区、皮质区Trk-B信号调节激酶表达的影响.方法:端粒酶敲除小鼠按随机数字表法分为手针组、电针组、空白对照组各6只.电针组使用一对华佗牌不锈钢针具(0.3 mm ×0.25 mm)对双侧足三里进行针刺干预,交替频率,强度0.5~1.2 mA,幅度2/100 HZ,每日30 min,连续干预7d.手针组使用华佗牌不锈钢针具(0.3 mm ×0.25 mm)对双侧足三里进行针刺干预,每5 min捻转1次,每次30 min,空白对照组不做任何干预.采用Western blot法检测大脑海马和皮质区域Trk-B的表达情况.结果:手针组小鼠在干预后自主活动程度较空白组均有所增加,海马区电针组与空白组对照组比较Trk-B表达明显增高;手针组与空白对照组比较Trk-B表达明显增高;但在皮质区电针组与手针组比较Trk-B表达明显降低;电针组与空白对照组比较表达明显降低.结论:电针对不同脑区Trk-B表达的影响不同,电针可促进海马区Trk-B的表达,相同的穴位刺激电针和手针的作用机制不同,可能与酪氨酸B(Trk-B)信号调节激酶表达有关.
