猪核线粒体假基因的分布特征研究

         

摘要

Nuclear mitochondrial pseudogenes (NUMT) have been found in many species. However, NUMT in pig a species of great economic importance, have not been reported. By blast searching mitochondrial genomic sequences in pig chromosomal genome database, 132 paralogous sequences were identified. These sequences ranged from 37 bps to 4453 bps in size, with 90 percent between 40 bps to 1000 bps, and the identity ranged from 66% to 100%. Pig NUMT were located on chromosomes 1, 4, 5, 7, 11, 13, 14, 15, and 17, covering total mitochondrial genome including all mtDNA coding regions and non-coding regions (the control region), and approximately half of them are located on chromosome 14. Furthermore, 30 complete mitochondrial genes associated with NUMT were discovered. These investigations provided valuable information for further study on mitochondrial DNA identification and phylogenetic analyses on pigs.%核线粒体假基因(NUMT)是线粒体DNA插入到核基因组中的DNA片段.目前,已经发现越来越多的真核生物基因组存在线粒体假基因现象.猪线粒体假基因在核基因组中的分布尚未见报道.研究通过猪线粒体基因组全序列与核基因组全序列进行比对,由BLAST程序鉴别出132个NUMT.结果表明:这些NUMT的大小在37~4 453bp,其中90%的NUMT长度处于40~1 000 bp的范围内,NUMT与相应的线粒体DNA片段之间的同源性数值范围为66%~100%.此外,鉴定出的NUMT序列几乎涵盖了包括线粒体DNA控制区在内的全部线粒体基因组,包括30个完整的线粒体基因,分布于猪的1、4、5、7、11、13、14、15、17号染色体上,近50%的NUMT定位在14号染色体上.
