重组枯草芽孢杆菌SE1对肉鸡生长性能、肠道消化酶活性和菌群的影响

     

摘要

本试验旨在研究芽孢表面展示鸡白痢沙门氏菌OmpC重组枯草芽孢杆菌SE1对肉鸡生长性能、肠道消化酶活性和菌群的影响.选择7日龄肉鸡120只,随机分为3组,每组4个重复,每个重复10只鸡.A组(对照组)饲喂基础饲粮,B组和C组分别在基础饲粮中添加0.1%(1.0×106CFU/g)枯草芽孢杆菌168制剂和0.1%(1.0×106CFU/g)重组枯草芽孢杆菌SE1制剂,试验期35 d.结果显示:1)与A组相比,C组肉鸡的终末体重和平均增重分别增加6.14%和6.76%(P>0.05),料重比降低8.21%(P>0.05).2)28、42日龄时,B组和C组肉鸡的空肠脂肪酶和回肠蛋白酶活性显著高于A组(P<0.05);B组和C组肉鸡的肠道消化酶活性无显著差异(P>0.05).3)28、42日龄时,B组和C组肉鸡的回肠、盲肠大肠杆菌数量显著低于 A 组(P<0.05),盲肠乳杆菌数量显著高于A组(P<0.05).16S rRNA V3区聚合酶链式反应-变性梯度凝胶电泳(PCR?DGGE)结果显示,B组和C组肉鸡的盲肠菌群丰富度、均匀度和香农-维纳指数均显著高于A组(P<0.05);C组肉鸡盲肠菌群的相似性在28、42日龄时分别比A组高26.8%和15.6%,C组和B组之间无显著差异(P>0.05).结果表明,重组枯草芽孢杆菌SE1具有与枯草芽孢杆菌168相同的效果,能有效促进肉鸡生长,提高肠道脂肪酶和蛋白酶活性,调节肉鸡肠道菌群,提高肠道菌群稳定性和多样性.%This experiment was conducted to investigate the effects of recombinant Bacillus subtilis SE1 spore surface display of Salmonella pullorum OmpC on growth performance,intestinal digestive enzyme activities and microflora of broilers. One hundred and twenty seven?day?old broilers were randomly divided into 3 groups with 4 replicates per group and 10 broilers per replicate. Broilers in group A(control group)were fed a basal diet, and the others in groups B and C were fed the basal diets supplemented with 0.1%(1.0×106CFU/g) Bacillus subtilis 168 and 0.1%(1.0×106CFU/g)recombinant Bacillus subtilis SE1,respectively. The experi?ment lasted for 35 days. The results showed as follows:1)compared with group A,the final weight and aver?age gain of broilers in group C increased by 6.14% and 6.76%(P>0.05), respectively, and ratio of feed to gain in group C decreased by 8.21%(P>0.05).2)The activities of jejunal lipase and ileal protease of broilers in groups B and C were significantly higher than those in group A at 28 and 42 days of age(P<0.05), and there were no significant differences on intestinal digestive enzyme activities of broilers between group B and group C(P>0.05). 3)At 28 and 42 days of age,the number of Escherichia coli in ileum and cecum of broil?ers in groups B and C was significantly lower than that in group A(P<0.05),and the number of Lactobacillus in cecum in groups B and C was significantly higher than that in group A(P<0.05). The results of polymerase chain reaction?denatured gradient gel electrophoresis(PCR?DGGE)of 16S rRNA V3 zone showed that the richness, evenness and Shannon?Wiener index of caecal microflora of broilers in groups B and C were signifi?cantly higher than those in group A(P<0.05), the similarity of caecal microflora in group C increased by 26.8% and 15.6% compared with group A at 28 and 42 days of age,respectively,and there was no significant difference between group C and group B(P>0.05). It is concluded that recombinant Bacillus subtilis SE1 have the same effect as Bacillus subtilis 168,can effectively promote growth of broilers,improve the activities of in?testinal lipase and protease,regulate the intestinal microflora of broilers,and improve the stability and diversity of intestinal microflora.
