氯化钴和跑台运动对大鼠海马神经发生及低氧诱导因子-1水平的影响

     

摘要

目的:比较氯化钴(CoCl2)所致机体低氧和跑台运动对大鼠海马齿状回新生细胞数量和低氧诱导因子-1(HIF-1)水平的影响.方法:腹腔注射CoCl2和BrdU于动物,运动方式为跑台运动,大鼠海马齿状回新生细胞和HIF-1采用免疫荧光显色显示.结果:小强度跑台运动组大鼠海马齿状回内BrdU免疫阳性(BrdU+)细胞数量显著高于对照组动物;使用CoCl2并进行小强度运动的动物,其海马齿状回内BrdU+细胞数也明显高于对照组动物;CoCl2组动物海马齿状回内BrdU+细胞数与对照组动物相比无显著性差异.小强度运动组动物、大强度运动组动物和CoCl2组动物海马齿状回内HIF-1水平皆高于对照组动物.结论:小强度跑台运动可以显著促进海马神经发生;跑台运动和CoCl2对海马的神经发生可产生叠加作用;CoCl2对大鼠海马齿状回的神经发生有一定促进作用,但无统计学意义.CoCl2和运动都可以导致海马齿状回HIF-1水平一定程度的增加.%Objective: To investigate the effects of CoClz and treadmill running on hippocampal neurogensis and hypoxia-in-ducible factor-1 (HIF-1) expression in young SD rats. Methods: Animals received intraperitoneal injections of BrdU and C0Cl2. Treadmill running was chosen for rat exercise. Immunofluorescence labeling was used to check the number of the newborn cells and the level of HIF-1 expression. Results: The numbers of BrdU positive (BrdU+ ) cells in hippocampal dentate gyrus of low-intensity exercise group and low-intensity exercise and C0Cl2 treated group were higher than that of the control group; significant difference was not found in the number of BrdU+ cells between CoCl2 treated and control group. The levels of HIF-1 in hippocampal dentate gyrus of low-intensity group, high-intensity group and C0Cl2 treated group were higher than that of the control group. Conclusion: The low-intensity treadmill running can obviously promote hippocampal neurogenesis in SD rats; the synergy of treadmill running and C0Cl2 on hippocampal neurogenesis is observed;although the enhancing trend of C0Cl2 treatment on neurogenesis was observed in SD rats, no obvious difference exists compared with the control group. Both C0Cl2 and exercise can induce HIF-1 expression in hippocampal dentate gyrus of SD rats.
