电针对帕金森病模型大鼠的作用

     

摘要

Objective: To explore the effect of electroacupuncture (EA) with different frequency in different time course on postural asymmetry and expression of tyrosine hydroxylase in substantia nigra pars (SNc) of Parkinson's disease (PD) model rats. Methods: Sprague-Dawley rats were randomly divided into normal control, sham, model, Madopar treatment, low-frequency and high-frequency EA groups. According to treatment course (1,2 and 3 weeks), the later five groups were subdivided into three subgroups. The PD model was duplicated by injecting 6-hydroxyl-dopamine (6-OHDA) into the right stria-tum of rats. And, the rats in EA groups were given low-frequency (2 Hz) and high-frequency (100 Hz) at "Hegu" and "Taichong" points. The postural asymmetry was observed, and the expression of TH in SNc was detected immunohisto-chemially. Results:High-frequency EA suppressed the increased rotation frequency to the right side that was showed in model rats. Additionally, high-frequency EA caused an up-regulation of TH expression in SNc, which was reduced in PD models. However, these effects were not observed in animals treated with low-frequency EA. Conclusion: This finding suggest that high-frequency EA has a therapeutic effect in the PD rats, attributing to its capability to reduce the rotational frequency to the lesioned side and up-regulate the TH expression in SNc.%目的:研究不同频率和疗程电针对帕金森病(PD)模型大鼠姿势不对称性和黑质致密部酪氨酸羟化酶(TH)表达的影响.方法:SD大鼠随机分为16组:正常对照组,1、2、3周假手术组、模型组,1、2、3疗程美多巴组、低频电针组、高频电针组,采用右侧纹状体内注射6-羟基多巴胺(6-OHDA)制备PD模型,取合谷和太冲穴,分别给予低频(2 Hz)和高频(100Hz)电针治疗.行为学观察大鼠姿势不对称性,免疫组织化学方法观察TH表达.结果:PD模型大鼠向右侧转动频率增加,高频电针可降低其向右侧转动频率,低频电针对其没有影响.PD模型大鼠黑质致密部的TH表达减少,高频电针可增加其TH表达,低频电针对其没有影响.结论:高频电针可降低PD模型大鼠向损毁侧转动频率,增加其黑质致密部的TH表达,提示高频电针对PD模型大鼠可能有治疗作用.
