移植骨髓间质干细胞在损伤脊髓内向神经元的定向分化

         

摘要

Objective:To explore the possibility that implanted bone marrow mesenchymal stem cells (BMSCs) can differentiate into neurons within injured spinal cord in rats. Methods:The BMSCs were implanted into the injured spinal cord of rats, and then underwent ultrastructural observation using immuno-electron microscopy, and immunohistochemical analysis against ChAT, GAD and synapsins on postsurgical week 1, 3 and 5. Results:One week after implantation, the BMSCs migrated into the grey matter of the spinal cord of rats with undeveloped organelles. The BMSCs did not exhibit the ultrastructural feathers of neuron, and negative immunoreactivity against ChAT , GAD and synapsins. Three weeks later, the implanted BMSCs within grey matter of the spinal cord of rats had developed organelles, ultrastructural feathers of neuron, and positive immunoreactivity against ChAT, GAD and synapsins. Conclusion:The implanted BMSCs within the injured spinal cord of rats can differentiate partly into functional neurons.%目的:探讨移植骨髓间质干细胞(BMSCs)在损伤脊髓内分化为功能性神经元的可能性.方法:应用低温包埋免疫电镜技术观察迁移在损伤脊髓内1、3、5周后的移植BMSCs的超微结构,应用免疫荧光标记和激光共聚焦技术观察迁移在损伤脊髓内1、3、5周后的移植BMSCs表达胆碱乙酰基转移酶(ChAT)、谷氨酸脱羧酶(GAD)和突触素(synapsins)的情况.结果:移植1周后,迁移在损伤脊髓灰质内的BMSCs细胞器不发达,不具有神经元的超微结构特点.移植3周后,迁移在损伤脊髓灰质内后的BMSCs细胞器开始发达,具有神经元的超微结构特点.移植1周后,迁移在损伤脊髓灰质内后的BMSCs不表达ChAT、GAD和突触素.移植3周后,迁移在损伤脊髓灰质内的BMSCs开始表达ChAT、GAD和突触素.结论:移植BMSCs在损伤脊髓内可部分分化为功能性神经元.
