基因芯片技术在阴道分泌物常规检测中的应用研究

摘要

目的:探讨基因芯片技术在阴道分泌物常规检测中的应用价值。方法选取2012年1-4月首都医科大学附属北京天坛医院妇产科门诊400例患者阴道分泌物,每份标本同时用 BD Affirm VPⅢ芯片检测、显微镜镜检、细菌性阴道病(BV)甲酚蓝试剂检测和 BV 蓝试剂检测,并接种至念珠菌显色培养基和滴虫培养基培养。采用 SPSS 16.0统计软件,行 Kappa 检验。结果以 BV 蓝试剂检测结果为金标准,BV 甲酚蓝试剂检测和 BD Affirm VPⅢ芯片检测 BV的 Kappa 值分别为0.813和0.553。以念珠菌培养结果为金标准,显微镜镜检和 BD Affirm VPⅢ芯片检测念珠菌的 Kap-pa 值分别为0.605和0.941。以滴虫培养结果为金标准,显微镜镜检和 BD Affirm VPⅢ芯片检测滴虫的 Kappa 值分别为0.973和0.923。结论 BD Affirm VPⅢ芯片检测念珠菌和滴虫一致性较好,BD Affirm VPⅢ芯片与 BV 蓝或 BV 甲酚蓝试剂联合用于临床进行阴道分泌物检查,可以取代显微镜镜检。%Objective To study the application of gene chip technique in leucorrhea routine examination. Methods Vaginal secretions of 400 gynecological outpatients referred to our hospital from January 2012 to April 2012 were tested with BD Affirm VPⅢ chips,microscopes,BV cresol blue agent and BV blue agent for testing bacterial vaginopathy respectively,which were cultured in candida color medium and trichomonad culture mediums. Kappa tests were performed with the statistical program SPSS 16. 0. Results The results of BV blue reagent were regarded as golden standard for testing bacterial vaginopathy,the Kap-pa values of BV cresol blue reagent and BD Affirm VPⅢ chips were 0. 813 and 0. 553 respectively. The results of candida chromo-genic plates were regarded as golden standard for testing candida,the Kappa values of microscope and BD Affirm VPⅢ chips were 0. 605 and 0. 941 respectively. The results of trichomonad culture mediums were regarded as golden standard for testing trich-omonad,the Kappa values of microscope and BD Affirm VPⅢ chips were 0. 973 and 0. 923 respectively. Conclusion The BD Affirm VPⅢ gene chips accompanied with BV blue reagent or BV cresol blue reagent can take the place of microscope examination to test vaginal discharge clinically.
