磷脂酶 A2-ⅡA 亚型基因 rs11677C/T 多态位点基因突变及蛋白表达与食管鳞状细胞癌临床病理特征的关联性研究

摘要

目的：探讨磷脂酶 A2-ⅡA 亚型（PLA2G2A）基因中的单核苷酸多态性（SNP）rs11677 C/ T 多态位点基因突变及蛋白表达与食管鳞状细胞癌（ESCC）临床病理特征的关系。方法选取2009年10月—2010年9月河南省林州市食管癌高发区，首次经组织病理学确诊的食管 ESCC 患者260例，血液标本采用 MassArray 时间飞行质谱生物芯片检测 PLA2G2A 基因 rs11677 C/ T 位点基因型；采用免疫组织化学染色方法（ SP 法）检测癌及癌旁组织中PLA2G2A 蛋白的表达情况。比较不同临床特征食管 ESCC 患者 PLA2G2A 基因 rs11677 C/ T 阳性率及 PLA2G2A 蛋白表达阳性率，不同 rs11677 C/ T 基因型食管 ESCC 患者 PLA2G2A 蛋白表达阳性率。结果260例食管 ESCC 患者中 PLA2G2A基因 rs11677 C/ T 多态位点 CC 型66.2%（172/260）、CT ＋ TT 型33.8%（88/260）。携带 CC 基因型者 PLA2G2A 蛋白表达阳性率（56.4%，97/172）与携带 CT ＋ TT 基因型者（55.7%，49/88）比较，差异无统计学意义（ P ＞0.05）。不同年龄、组织学分化、TNM 分期、淋巴结转移、浸润深度食管 ESCC 患者 PLA2G2A 基因 rs11677 C/ T 基因型频率比较，差异均无统计学意义（P ＞0.05）。不同年龄、组织学分化、TNM 分期、淋巴结转移食管 ESCC 患者 PLA2G2A 蛋白表达阳性率比较，差异均无统计学意义（P ＞0.05）；不同浸润深度食管 ESCC 患者 PLA2G2A 蛋白表达阳性率比较，差异有统计学意义（P ＜0.05）。不同 rs11677C/ T 基因型食管 ESCC 患者 PLA2G2A 蛋白表达阳性率比较，差异无统计学意义（χ2＝0.149，P ＝0.928）。结论未发现 PLA2G2A 基因 rs11677 C/ T 多态位点基因突变与其蛋白表达的相关性，PLA2G2A 蛋白表达与食管 ESCC 浸润深度相关。%Objective To investigate the correlation among the phospholipase A2 group ⅡA(PLA2G2A)gene single nucleotide polymorphism(SNP)(rs11677 C/ T),the protein expression and the clinicopathologic characteristics of esophageal squamous cell carcinoma(ESCC). Methods A total of 260 ESCC patients from Linzhou city,Henan province,an area of high incidence of esophageal cancer,were selected as study subjects from October 2009 to September 2010,ESCC was confirmed by histopathologic examination for the first time. Blood specimens were used to detect the PLA2G2A rs11677 C/ T SNP genotype by time - of - flight mass spectrometry MassArray system,and the carcinoma and pericarcinous tissues were used to examine the expression of PLA2G2A protein by immunohistochemical SP method. The positive rates of PLA2G2A rs11677 C/ T and PLA2G2A protein expression were compared among different groups of ESCC patients with different clinical features,the positive rates of PLA2G2A protein expression were compared among different groups of ESCC patients with different rs11677 C/ T genotypes. Results Among 260 ESCC patients,the distribution of CC,TT + CT genotype of PLA2G2A rs11677 C/ T SNP was 66. 2% (172 / 260)and 33. 8% (88 / 260),respectively. There was no significant difference in the positive rate of PLA2G2A protein expression between cases with CC genotype(56. 4% ,97 / 172)and cases with CT + TT genotype(55. 7% ,49 / 88)(P> 0. 05). The genotypes rate of PLA2G2A rs11677 C/ T did not vary by age,histological differentiation,TNM stage,lymphatic metastasis and depth of invasion of ESCC(P > 0. 05). The positive rate of PLA2G2A protein expression did not vary by age, histological differentiation,TNM stage and lymphatic metastasis(P > 0. 05). There was no significant difference in the positive rate of PLA2G2A protein expression among ESCC patient groups with different rs11677 C/ T genotypes( χ2 = 0. 149,P =0. 928). Conclusion The correlation between PLA2G2A rs11677 C/ T polymorphism and the PLA2G2A protein expression is not significant,while PLA2G2A protein expression is closely related to the depth of invasion of ESCC.
