复方中药对链球菌性肺炎小鼠肺组织炎性细胞因子的影响研究

摘要

目的:探讨复方中药对链球菌性肺炎小鼠肺组织炎性细胞因子白介素( IL)-6、IL-2、干扰素γ(INF-γ)、肿瘤坏死因子α(TNF-α)、IL-10表达水平的影响。方法2014年3—10月选取健康 BALB/ c 小鼠36只,采用随机数字表法分为3组:正常组、链球菌性肺炎模型组(模型组)、复方中药治疗组(中药组),每组12只。模型组、中药组小鼠分别破损鼻黏膜,滴入50μl 配好的肺炎链球菌标准菌株溶液;正常组小鼠破损鼻黏膜,滴入50μl 无菌0.9%氯化钠溶液。中药组小鼠给予7.5 g·kg -1·d -1复方中药灌胃,正常组和模型组小鼠给予等剂量双蒸水灌胃,均2次/ d,连续给药7 d。给药第7天,取肺组织,观察病理学表现,酶联免疫吸附试验(ELISA)法检测各组小鼠肺组织IL-6、IL-2、INF-γ、TNF-α、IL-10表达水平。结果苏木素-伊红(HE)染色光镜下显示,正常组肺泡结构及上皮完整,肺泡腔清晰,肺泡内无渗出物,肺泡间隔均匀一致,无增厚,毛细血管无明显扩张、充血及炎性细胞浸润;模型组肺泡腔内充满渗出的纤维素,出现大量炎性细胞浸润,肺泡间隔增厚及毛细血管扩张、充血;中药组肺泡腔内纤维素渗出较少,肺间质有少量炎性细胞浸润,少量红细胞,部分肺泡破坏融合,与模型组比较有所减轻。模型组小鼠肺组织IL-6、IL-2、INF-γ、TNF-α、IL-10表达水平较正常组升高(P <0.05);中药组小鼠肺组织IL-6、IL-2、INF-γ、TNF-α、IL-10表达水平较模型组降低(P <0.05);中药组小鼠肺组织IL-6、IL-2、INF-γ、TNF-α、IL-10表达水平与正常组比较,差异无统计学意义(P >0.05)。结论复方中药能有效抑制链球菌性肺炎小鼠肺组织IL-6、IL-2、INF-γ、TNF-α、IL-10的过度表达,缓解炎性反应。%Objective To investigate the effects of complex prescription of traditional Chinese medicine on expression levels of IL-6,IL-2,INF-γ,TNF-α,and IL-10 in lung tissue of streptococcus pneumonia mice. Methods From March to October in 2014,we selected 36 healthy BALB/ c mice and used random number table method to divide them into 3 groups, namely control group,streptococcus pneumonia model group( model group),and the group of complex prescription of TCM (TCM group),with 12 mice in each group. The mice in the model group and TCM group were given 50 μl standard solution of streptococcus pneumoniae through damaged nasal mucosa,and the mice in control group were given the same mount of 0. 9%sodium chloride liquid. Mice in TCM group was given complex prescription of TCM by gavage by 7. 5 g·kg - 1 ·d - 1 ,and control group and model group were given the same amount of double distilled water by 2 times per day for 7 days consecutively. Lung tissues were taken in the 7 dany to observe pathological manifestations,and ELISA method was used to detect the expression levels of IL-6,IL-2,INF-γ,TNF-α and IL-10 in lung tissue. Results After the observation by HE staining and light microscope,we found the following results. In control group,the alveolar structure and epithelium were complete with clear alveolar space and no transudation;the alveolar space was homogeneous without incrassation;no obvious expansion,congestion and inflammatory cell infiltration occurred in inner capillaries. In model group,alveolar space was full of cellulose exudate;plenty of inflammatory cell infiltration occurred; alveolar space had incrassation; expansion and congestion occurred in capillaries. In TCM group,there was little cellulose exudate in alveolar space,little inflammatory cell infiltration in pulmonary mesenchyme and a small number of red blood cells;a part of pulmonary alveolis were damaged and merged;the condition was much relieved compared with model group. Model group was higher than control group in the expression levels of IL-6,IL-2, INF-γ,TNF-α and IL-10 in lung tissue(P < 0. 05). TCM group was lower than model group in the expression levels of IL-6, IL-2,INF-γ,TNF-α and IL-10 in lung tissue(P < 0. 05);TCM group was not significantly different from control group in the expression levels of IL-6,IL-2,INF-γ,TNF-α and IL-10 in lung tissue( P > 0. 05). Conclusion Complex prescription of TCM can effectively inhibit the excessive secretion of IL-6,IL-2,INF-γ,TNF-α,and IL-10 in lung tissue of streptococcus pneumonia mice,and relieve inflammatory reaction.
