新生鼠视网膜中TGF-β1表达的影响研究

     

摘要

AIM: To compare the expression of the TGF-β1 in the retina of different oxygen inhalation mice and reveal the relation between TGF-β1 and retinopathy of prematurity. METHODS:A total of 60 C57BL /6J mice were randomly assigned into 3 groups. The mice were raised in atmosphere in the control group. Experimental group 1 was received 75% of oxygen for continuous 5 days. Experimental group 2 was received 30%,40%,50%,60%,75%,60%,50%,40%,30%,20% oxygen in turn for one day respectively. The retinal vascular development were evaluated by the retinal flat-mounts with ADP stained at 3,8 and 14 day. The expressions of TGF-β1 and TGF-β1 mRNA in each group were detected with immunohistochemical method and RT-PCR at 14 day. RESULTS: The retinal vessels of experimental group 1 and 2 was significantly thinner in comparison with control group. At 14 day, the density of peripheral retinal vascular of experimental group 1 were higher than experimental group 2 and control group. The expression of TGF-β1 and TGF-β1 mRNA of experimental group 1 was significantly higher than that in experimental group 2 and control group. CONCLUSION: The TGF-β1 was related to the pathogenesy of retinopathy of prematurity. The obvious fluctuation of inhaled oxygen may severely affect the expression of TGF-β1 and TGF-β1 mRNA, and lead to the pathologic changes similar to retinopathy of prematurity.%目的比较TGF-β1在不同氧环境饲养新生鼠视网膜中的表达变化,揭示TGF-β1与早产儿视网膜病变发病的关系.方法60只7日龄C57BL/6J鼠仔随机分为三组,对照组饲养于空气中;实验组1饲养于75%氧箱中5天后改为空气饲养;实验组2氧箱中的氧浓度逐日从30%升高至75%后再逐日下降至空气饲养.分别于第3、8、14天行视网膜铺片ADP染色观察视网膜血管变化;14天时采用免疫组化法检测视网膜TGF-β1表达,RT-PCR法检测TGF-β1 mRNA表达.结果实验组1和实验组2新生鼠视网膜血管缩窄.第14天时,实验组1周边视网膜血管密度增加,TGF-β1表达呈阳性改变,TGF-β1 mRNA表达高于对照组和实验组2(P>0.05);实验组2周边视网膜血管未见明显异常改变,TGF-β1表达呈阴性改变,TGF-β1 mRNA表达与对照组比较差异无统计学意义(P<0.05).结论 TGF-β1参与早产儿视网膜病变的发病过程,吸入氧浓度波动较大时视网膜表达TGF-β1增加,并产生类似于早产儿视网膜病变的改变.
