目的:观察应用RNAi技术下调局部粘着斑激酶(FAK)基因表达对SW620细胞侵袭及转移能力的影响。方法利用前期构建的针对FAK基因有效的短发夹RNA (shRNA )序列的慢病毒表达载体,包装慢病毒并感染人结肠癌细胞SW620。运用免疫印记(Western-blot)从蛋白水平检测FAK基因的表达, Transwell小室模型分别检测敲减FAK基因表达对SW620细胞的侵袭及转移能力的影响。结果用慢病毒敲减FAK后的大肠癌细胞,蛋白水平FAK表达明显下调, SW60细胞的侵袭及转移能力均受到明显抑制。结论下调FAK的表达水平,能明显抑制大肠癌SW620细胞的侵袭及转移能力。%ObjectiveTo observe influence by RNAi down-regulated focal adhesion leinase (FAK) gene on invasion and metastasis ability in colorectal cancer cell SW620.MethodsLentivirus packaging was made by expression vector of short hairpin RNA (shRNA) sequence established for FAK gene, and human colon cancer cell SW620 was infected. Western-blot was applied to detect FAK gene expression from protein level, and Transwell cell model was used to detected influence by FAK gene knock-down expression on invasion and metastasis ability in SW620.ResultsIn colorectal cancer cell after FAK gene knock-down by lentivirus, protein FAK expression was reduced, while invasion and metastasis ability in SW620 were obviously suppressed.ConclusionInvasion and metastasis ability in colorectal cancer cell SW620 can be remarkably suppressed by down-regulated FAK expression.
