OBJECTIVE: To establish analysis method of HPLC specific chromatograms of crude and processed Gansu Angelica sinensis, and to determine the content of ferulic acid before and after processing. METHODS: The HPLC separation was performed on Agilent HC-C18(250 mm×4.6 mm, 5 μm) column by gradient elution using acetonitrile-0.2% aqueous acetic acid as mobile phase at the flow rate of 1.0 mL-min-1. The column temperature was 35 ℃ and the detection wavelength was set at 254 nm. RESULTS: Specific chromatograms of Gansu A. sinensis from 3 main producing areas showed high similarity. The content of ferulic acid decreased a little in processed Gansu A. sinensis. CONCLUSION: Specific chromatograms combined with marker compounds determination can be used for quality control of Chinese medicinal materials and Chinese herbal pieces. This method shows high precision, good repeatability and contents are separation. It can be used for the quality evaluation of crude and processed A sinensis.%目的:建立甘肃当归生品和炮制品的高效液相色谱(HPLC)特征图谱分析方法,同时测定炮制前后当归中阿魏酸的含量.方法:色谱柱为Agilent HC-C18(250 mm×4.6 mm,5μm),流动相为乙腈-0.2%乙酸水溶液(梯度洗脱),检测波长为254 nm,流速为1.0 mL·min-1,柱温为35℃.结果:3个主产地的甘肃当归HPLC特征图谱相似度较高;炮制后阿魏酸的含量略有降低.结论:采用特征图谱技术结合指标性成分含量测定的方法可以较好地控制药材及饮片的质量.该法精密度高、重复性好,所测组分均达到基线分离,可以用于炮制前后当归的质量评价.
