半夏5种不同溶剂提取物对小鼠祛痰镇咳作用的研究

         

摘要

OBJECTIVE: To study cough relieving and sputum removing effect of 5 different solvent extracts of Pinellia ternata on mice. METHODS: The sputum removing effect was evaluated with phenol red excretion. 70 kunming mice were divided into blank control group (constant volume of normal saline), ammonium chloride(0.5 g·kg-1) group, alcohol extract off! Ternate(2 g·kg-1) group, water extract of P. Temate(1.2 g·kg-1) group, petroleum ether extract of P. Ternate(1.2 g·kg-1) group, ethyl acetate extract of P. Ternate(1.2 g·kg-1) group, butanol extract of P. Ternate (1.2 g·kg-1) group. All groups were given medicine intrarngastrically once a day for consecutive 7 days except for ammonia chloride group. Ammonia chloride group was given medicine once on 7th day.The cough relieving effect was evaluated with ammonia water induced cough model. 70 kunming mice were divided into model control group (constant volume of normal saline), codeine(0.002 g·kg-1) group, alcohol extract of P. Ternate(0.6 g·kg-1) group, water extract of P. Ternate(0.6 g·kg-1) group, petroleum ether extract of P. Ternatei 0.6 g·kg-1) group, ethyl acetate extract of P. Ternate(0.6 g·kg-1) group, butanol extract of P. Ternate(0.6 g·kg-1) group. All groups were given medicine intragastrically once a day for consecutive 7 days except for codeine group. Ammonia chloride group was given medicine once on 7th day. The incubation period of cough and cough frequency in 2 min. RESULTS: Compared with the model control group, phenol red excretion of each treatment group increased significantly(P<0.01 or P<0.05), especially butanol extract of P. Ternate; Compared with the model control group, the incubation period of cough in each treatment group were obviously prolonged, while cough frequency was significantly reduced in 2 min(P<0.01 or P<0.05), especially butanol extract of P. Ternate. CONCLUSION: Among 5 different solvent extracts of f! Ternata, butanol extract show the best cough relieving and sputum removing effect, which can provide reference for the clinic.%目地:研究半夏5种不同溶剂提取物对小鼠的镇咳、祛痰作用.方法:以酚红排泄量法评价祛痰作用.70只昆明种小鼠分为空白对照(等体积生理盐水)、氯化铵(0.5 g·kg-1、半夏醇提物(1.2 g·kg-1)、半夏水提物(1.2 g·kg-1)、半夏石油醚提取物(1.2g·kg-1)、半夏乙酸乙酯提取物(1.2g·kg-1)、半夏正丁醇提取物(1.2 g·kg-1)组.灌胃给药,每天1次,除氯化铵组外其余各组连续7d,氯化铵组第7天给药1次.以氨水引咳法评价镇咳作用.70只昆明种小鼠分为模型对照(等体积生理盐水)、可待因(0.002 g·kg-1)、半夏醇提物(0.6 g·kg-1)、半夏水提物(0.6 g·kg-1)、半夏石油醚提取物(0.6 g·kg-1)、半夏乙酸乙酯提取物(0.6 g·kg-1)、半夏正丁醇提取物(0.6 g·kg-1)组.灌胃给药,每天1次,除可待因组外其余各组连续7d,可待因组第7天给药1次.结果:与空白对照组比较,各用药组酚红排泄量显著增加(P<0.01或P<0.05),其中半夏正丁醇提取物作用最强.与模型对照组比较,各用药组小鼠咳嗽潜伏期显著延长,2 min内咳嗽次数显著减少(P<0.01或P<0.05),其中半夏正丁醇提取物效果最好.结论:半夏5种不同溶剂提取物中正丁醇提取物对小鼠的祛痰、镇咳作用最好.
