Objective To establish an UPLC method for content determination of trepibutone. Methods The Waters Atlantis T3 column (100 mm × 2. 1 mm, 3 μm) was adopted, the mobile phase was 0. 01 mol/L sodium acetate (adjusted p H to 3. 0 by phosphoric acid) -methanol (40 ∶ 60), the flow rate was 0. 5 m L/min, and the column temperature was 35 ℃. DAD detector was used to detect, the qualitative analysis was carried out based on the retention time, and the content of trepibutone was determined at the wavelength of 271 nm.Results The linearity of peak area was good when the content of trepibutone was in the range of 0. 100 04-1. 000 40 g/L, r =0. 999 92 (n = 5), the mean recovery rate was 99. 89%, RSD was 0. 61% (n = 9). Conclusion This method is accurate and efficient, which can be used for the content determination of trepibutone.%目的 建立测定曲匹布通含量的超高效液相色谱(UPLC)法.方法 色谱柱采用Waters AtlantisT3柱(100 mm×2.1 mm,3μm),流动相为0.01 mol/L的醋酸钠溶液(用冰醋酸调节pH至3.0)-甲醇(40∶60),流速为0.5 m L/min,柱温为35℃,用DAD检测器检测,以保留时间定性,在271 nm波长下测定曲匹布通的含量.结果 曲匹布通质量浓度在0.100 04~1.000 40 g/L范围内与峰面积线性关系良好,r=0.999 92(n=5);平均回收率为99.89%,RSD为0.61%(n=9).结论 该方法准确、高效,可用于曲匹布通的含量测定.
