甘草对红花中羟基红花黄色素A药代动力学的影响

             

摘要

Objective Flos Carthami and Radix Glycyrrhizae were selected as example representing key synergistic relationship of complex prescription to elucidate the harmonizing effect and substantial foundation of Radix Glycyrrhizae in a formula by investigating the effect of Radix Glycyrrhizae on pharmacokinetic of hydroxysafflor yellow A, an active ingredient in Flos Carthami. Methods Trichloroacetic acid was used to remove plasma protein in the samples. Reverse phase high performance liquid chromatography with external standard method was employed to determine the concentration of hydroxysafflor yellow A. Software of DAS 2. 0 was employed to regress the drug - plasma concentration curve and calculate pharmacokinetic parameters of hydroxysafflor yellow A. Results The pharmacokinetic profiles of hydroxysafflor yellow A confirm to one and two compartment open model for groups of Radix Glycyrrhizae and Carthamus tinctorius L. Coupled with Radix Glycyrrhizae, respectively. The absorption half - life and elimination half - life of HSYA shortened significantly after compatibility. Meanwhile, the area under the curve (AUC) was significantly decreased. Conclusion Radix Glycyrrhizae exerts its harmonizing effect by enhancing absorption and elimination of hydroxysafflor yellow A.%目的 选择代表复方关键配伍关系的红花-甘草(君-使)药对为研究对象,通过探讨甘草对红花水提液中有效成分羟基红花黄色素A药代动力学的影响,揭示复方中使药调和作用的物质基础及其机制.方法 以三氯乙酸沉淀法去除血浆蛋白,反相高效液相色谱外标法测定羟基红花黄色素A的含量.以DAS 2.0软件拟合药时曲线,计算药代动力学参数.结果 配伍前后,羟基红花黄色素的药代动力学曲线分别符合二室开放模型和一室开放模型;配伍后,羟基红花黄色素A的吸收半衰期和消除半衰期明显缩短,药时曲线下面积明显减小.结论 甘草促进红花的吸收和消除为其发挥调和作用的机制之一.
