高山被孢霉原生质体菌龄的研究

             

摘要

The most available fungus age of Mortierella alpina protoplast was determined. Protoplasts of Mortierella alpina 200012201(1) with different fungus ages were prepared by Lywallzyme and Snail enzymes mixture enzymolysis liquid (2% +4%). After inoculation and continuous fermentation in a 5 L tank, the products were detected by dying with sudan black B and gas chromatography. The results showed that the content of arachidonic acid in the product was influenced by protoplasts of Mortierella alpina with different fungus ages; the most available fungus age was 12 h; after fermentation for 7 d, the content of total lipid and arachidonic acid were 17. 3 g/L and 7. 55 g/L, respectively.%确定了适合制备生产用高山被孢霉(Mortierella alpina)原生质体的最佳菌龄.将溶壁酶和蜗牛酶按比例(2%+4%)配制成混合酶解液制备不同菌龄Mortierella alpina 200012201(1)的原生质体,分别进行5L发酵罐连续发酵,通过苏丹黑B染色和气相色谱法测定发酵产物.结果表明:菌龄对高山被孢霉原生质体发酵产物中花生四烯酸的含量有一定影响,选择菌龄为12 h的高山被孢霉原生质体进行发酵培养较为理想,培养7d后总油脂和花生四烯酸含量分别为17.3 g/L和7.55 g/L.
