针刺对高脂血症小鼠心脏血管紧张素原和内皮素及其受体mRNA水平的影响

摘要

Objective To investigate the effect of acupuncture on mRNA levels of angiotensinogen (AGT),angiotensin Ⅱ type 1 receptor (AT1R),endothelin 1 (ET1) and endothelin receptor A (ETAR) in cardiac tissue of hyperlipidemia mice.Methods Totally 21 apolipoprotein E gene knockout (APOE-/-) male mice with 16 weeks of age were completely randomly divided into acupuncture acupoint group,acupuncture non-acupoint group and model control group,7 mice in each group.Another 7 C57BL/6 mice were selected as the normal control group.Mice in acupuncture acupoint group were acupunctured at Neiguan and Fenglong acupoints with reinforcing-reduction by entwisting method under the condition of tying up,30 s/time,3 times/week,lasted for 8 weeks;mice in acupuncture non-acupoint group were acupunctured at 2 points respectively 0.5 cm and 1.0 cm from the base of tail,horizontally inserting 5 mm with an angle of 15° to the tail base,with the same method and time as acupuncture acupoint group;model and normal control group were only tied up 3 times/week,30 s/time.Tail blood were taken and serum levels of total cholesterol (TC) were tested at the first day of the first week and ninth week.The hearts were resected at the second day of the ninth week,and the mRNA levels of AGT,AT1 R,ET1 and ETAR in heart tissues were tested by real-time fluorescence quantitative polymerase chain reaction.Results The serum levels of TC in the first and ninth weeks in acupuncture acupoint group,acupuncture non-acupoint group and model control group were all significantly higher than normal control group [(10.65 ±1.17),(10.67 ± 1.46),(10.86 ±1.39)mmol/L vs (2.72 ±0.29)mmol/L,(19.34 ±2.33),(25.44 ±1.97),(25.91 ± 2.87) mmol/L vs (2.36 ± 0.36) mmol/L],and the serum level of TC in the ninth week in acupuncture acupoint group was significantly lower than acupuncture non-acupoint group and model control group (all P < 0.05).The mRNA levels of AGT in 4 groups had no statistically significant differences (all P > 0.05).The mRNA levels of AT1R and ET1 in acupuncture acupoint group,acupuncture non-acupoint group and model control group were significantly higher than normal control group [AT1R mRNA:(0.001 71 ± 0.000 44),(0.002 37 ±0.000 74),(0.002 94 ±0.000 73) vs (0.001 28 ±0.000 77),ET1 mRNA:(0.000 230 ±0.000 040),(0.000 204 ±0.000 034),(0.000 324 ±0.000 090) vs (0.000 133 ±0.000 102)],and in acupuncture acupoint group were significantly lower than those in model control group(P < 0.05).The mRNA level of ETAR in acupuncture acupoint group was significantly lower than acupuncture non-acupoint group,model and normal control group [(0.001 92 ± 0.000 73) vs (0.005 06 ± 0.001 35),(0.006 20 ± 0.001 69),(0.002 08 ±0.000 98)],and the mRNA levels of ETAR in model control group and acupuncture non-acupoint group were significantly higher than normal control group (P < 0.05).Conclusion Acupuncture at Neiguan and Fenglong acupoints can inhibit the rising of blood lipid in ApoE-/-mice.Acupuncture at acupoints is more effective than non-acupoints,the decreasing of mRNA levels of ET1,AT1R and ETAR in cardiac tissue stimulated by acupuncture may be the related reason.%目的 探讨针刺对高脂血症小鼠心脏组织血管紧张素原(AGT)、血管紧张素Ⅱ受体1(AT1R)、内皮素1以及内皮素受体A(ETAR) mRNA水平的影响.方法 选择16周龄雄性载脂蛋白E基因敲除(ApoE-/-)小鼠21只,完全随机分为针刺穴位组、针刺非穴位组和模型对照组,每组7只,另选7只C57BL/6小鼠作为正常对照组.针刺穴位组小鼠捆绑下针刺内关和丰隆穴,行平补平泻捻转手法30 s,每周3次,共8周.针刺非穴位组小鼠捆绑下取距离尾根部0.5 cm和1.0 cm各一点,针尖向尾根部15°平刺入5 mm,针刺手法和时间同针刺穴位组.正常对照组和模型对照组小鼠每周捆绑3次,每次30 s,不针刺.分别于第1周第1天和第9周第1天断尾取血检测小鼠血清总胆固醇含量,于第9周第2天取小鼠心脏,采用实时荧光定量聚合酶链反应检测并比较小鼠心脏组织中AGT、AT1R和内皮素1、ETAR mRNA水平.结果 第1和第9周,针刺穴位组、针刺非穴位组和模型对照组小鼠血清总胆固醇水平均明显高于正常对照组[(10.65±1.17)、(10.67±1.46)、(10.86±1.39) mmol/L比(2.72±0.29) mmol/L,(19.34±2.33)、(25.44±1.97)、(25.91±2.87) mmol/L比(2.36 ±0.36) mmol/L],但第9周针刺穴位组血清总胆固醇水平明显低于针刺非穴位组和模型对照组,差异均有统计学意义(均P<0.05).4组AGT mRNA水平差异均无统计学意义(均P>0.05).针刺穴位组、针刺非穴位组和模型对照组小鼠AT1R、内皮素1 mRNA水平明显高于正常对照组[AT1R mRNA:(0.001 71 ±0.000 44)、(0.002 37±0.000 74)、(0.002 94 ±0.000 73)比(0.001 28±0.00077),内皮素1 mRNA:(0.000 230±0.000 040)、(0.000 204 ±0.000 034)、(0.000324±0.000090)比(0.000 133 ±0.000 102)],且针刺穴位组AT1R、内皮素1 mRNA水平均明显低于模型对照组,差异均有统计学意义(均P<0.05).针刺穴位组ETAR mRNA水平明显低于针刺非穴位组、模型对照组和正常对照组[(0.001 92 ±0.000 73)比(0.005 06±0.001 35)、(0.006 20 ±0.001 69)、(0.002 08±0.000 98)],且模型对照组和针刺非穴位组ETAR mRNA水平明显高于正常对照组,差异均有统计学意义(均P <0.05).结论 针刺内关和丰隆穴可以抑制ApoE-/-小鼠血脂升高,针刺穴位比非穴位作用更明显,可能与针刺穴位能抑制心脏局部内皮素1、AT1R和ETAR mRNA水平有关.
