丹红注射液预处理对大鼠缺血再灌注心肌细胞损伤的影响

     

摘要

目的 研究丹红注射液预处理对大鼠心肌细胞缺血再灌注损伤的作用.方法 体外培养大鼠心肌细胞,将丹红注射液以2%、4%、8%、16%四种浓度分别加入培养基中,预处理24 h 后,置于缺血再灌注(I/R)环境中培养,采用流式细胞仪技术检测各组细胞的凋亡率,免疫组织化学法检测凋亡基因Bax 及Bcl-2 蛋白表达变化.结果 与正常对照组比较,缺血再灌组细胞凋亡率及Bas 表达明显升高,Bcl-2 表达则显著降低;而用丹红注射液预处理后呈浓度依赖性地下调凋亡率及Bas 表达,上调Bcl-2 表达.结论 丹红注射液对缺血再灌心肌细胞有保护作用,且与浓度有一定的依赖关系,其作用可能是通过上调Bcl-2 蛋白表达,抑制Bax 蛋白表达实现的.%Objective To investigate the effect of Danhong Injection pretreatment on cardiomyocytes cell of ischemic - reperfusion rats. Methods The rats cardiomyocytes were cultured and pretreated with Danhong Injection in four different concentrations: 2%, 4%, 8%, 16% before create cardiomyocytes I/R model after 24 hours for pretreatment. The apoptotic rate of cardiomyocytes was determined by flow cytometry, the protein expressions of Bel-2 and Bax were observed by imn -mnohistochemistry. Results Compared with control group, the apoptotic rates were significantly increased in ischemia-reperfusion (I/R) group, and the protein expressions of Bax expressions was significantly up-regulated and Bcl-2 expres -sions was down-regulated. Pretreatment of cardiomyocytes with Danhong Injection increased the expression of Bel-2, de -creased the expression of Bax and the apoptotic rate in a dose-dependent fashion. Conclusion Danhong Injection alleviates I/R injury in a concentration-dependent manner. These effects may be partially that Danhong Injection can inhibit Bas gene protein expression and induce Bel-2 gene protein expression.
