土大黄根提取物对小鼠尾部鳞片角化不全模型的影响

         

摘要

目的:探讨土大黄根提取物对小鼠尾部鳞片角化不全模型的影响。方法将70只KM小鼠随机分为对照组、地塞米松组、消银颗粒组、异常黏液质成熟剂组、土大黄根提取物组(1 g/kg组、2 g/kg组、4 g/kg组)7个组,每组各10只,按体重分别给予0.5%羧甲基纤维素钠、1.05 mg/kg地塞米松、1.577 g/kg消银颗粒、2 g/kg异常黏液质成熟剂及1、2、4 g/kg土大黄根提取物灌胃。采用小鼠尾部鳞片角化不全模型,观测灌胃给药后小鼠的一般状况、体重、脏器指数、血生化指标及尾部皮肤组织病理学变化。结果与对照组比较,1、2、4 g/kg组小鼠行为活动、皮毛、粪便、饮食等均未见异常,脏器指数、体重无明显变化,差异无统计学意义(P>0.05);血清肌酐、葡萄糖、总蛋白、白蛋白、总胆固醇、高密度脂蛋白-胆固醇、低密度脂蛋白-胆固醇均升高(P<0.05或P<0.01)。与对照组比较,1 g/kg组血清碱性磷酸酶升高、磷酸肌酸激酶降低;2 g/kg组血清丙氨酸氨基转移酶、天门冬氨酸氨基转移酶升高,尿素降低;4 g/kg组血清丙氨酸氨基转移酶、天门冬氨酸氨基转移酶、碱性磷酸酶、三酰甘油升高,磷酸肌酸激酶、尿素降低,差异均有统计学意义(P<0.05或P<0.01)。1、2、4 g/kg组促进小鼠尾部鳞片表皮颗粒层形成,且呈剂量规律性。结论土大黄根提取物能改变银屑病表皮的角化不全,但其作用与血生化指标的变化可能无明显关联。%Objective To explore the effect of Rumex L. root extract on mouse tail model. Methods 70 mice were randomly assigned to CON group, DXM group, XYKL group, MB group and Rumex L.root extract group (1 g/kg group, 2 g/kg group, 4 g/kg group), with 10 mice in each group, and according to the weight they were given 0.5% CMC-Na, 1.05 mg/kg Dexamethasone, 1.577 g/kg Xiaoying granules, 2 g/kg Munziq Balgam, and 1, 2, 4 g/kg Rumex L.root extract respec-tively by gavage. After gavage, general condition, body weight, organ index, blood biochemistry, tail skin histopathology of mice were observed by mouse tail scales parakeratosis model. Results Compared with CON group, mice of the 1, 2, 4 g/kg group had no obvious abnormalities in activity, fur, faeces, diet, and had no obvious change in organ index and body weight, there were no statistically significant differences (P>0.05);serum Crea, GLU, TP, ALB, TCHO, HDL-C, LDL-C of the 1, 2, 4 g/kg group increased (P<0.05 or P<0.01). Compared with CON group, serum ALP of the 1 g/kg group increased and CK decreased; serum ALT, AST of the 2 g/kg group increased and Urea decreased; serum ALT, AST, ALP of the 4 g/kg group increased and CK, Urea decreased (P<0.05 or P<0.01). 1, 2, 4 g/kg group promoted formation of epidermis granular layer on mouse tail skin and dose dependent. Conclusion Rumex L.root extract can change parakeratosis of psoriasis skin, which has a therapeutic effect, but its function and blood biochemical parameters of mice may not have clear correlation.
