高效液相色谱法同时测定百华花楸果实中金丝桃苷和芦丁的含量

         

摘要

目的 建立同时测定百华花楸果实中金丝桃苷和芦丁含量的高效液相色谱分析方法.方法 采用高效液相色谱法,色谱柱为Zorbax SB-C18柱(5μm,4.6 mm×250 mm),流动相为甲醇:乙腈:0.025 mol/L磷酸(10:20:70),流速1.0 mL/min,检测波长363 nm,柱温30℃.结果 金丝桃苷和芦丁分别在0.1~0.8μg和0.16~1.28μg范围内,进样量与色谱峰面积呈良好线性关系,相关系数分别为0.9994和0.9992;平均回收率(n=9)分别为98.58%和98.22%,RSD分别为1.71%和1.81%;平均含量分别为0.473、0.342 mg/g.结论 该法操作简便、准确,稳定性及重复性好,可用于对百华花楸果实中金丝桃苷和芦丁成分的含量测定.%Objective To establish HPLC method for the determination of hyperoside and rutin in fruit of Srobus po-huashanensis. Methods The Zorbax SB-C18 (5μm, 4.6 mm×250 mm) column was adopted and eluted with mobile phase consisted of methanol:acetonitrile:0.025 mol/L aqueous phosphoic acid (10:20:70) at the column temperature of 30℃. The flow rate was 1.0 mL/min and detective wave length was 363 nm. Results Hyperoside was proved to be linear in the range of 0.16-1.28 μg with a regression coefficient of 0.9992, the average recovery was 98.22% with RSD of 1.71%; Rutin was proved to be linear in the range of 0.1-0.8 μg with a regression coefficient of 0.9994, the average recovery was 98.58% with RSD of 1.81%. The average content of hyperoside and rutin was 0.473 mg/g and 0.473 mg/g respectively. Conclusion The developed method is convenient and accurate with high repeatability and stability, which is suitable for the content determination.
