黄诺玛苷对高糖诱导HUVEC细胞氧化应激及VEGF和ICAM-1蛋白表达的影响

         

摘要

目的 观察两色金鸡菊黄酮类化合物黄诺玛苷对高糖诱导的人脐静脉内皮细胞(HUVEC)氧化应激及血管内皮生长因子(VEGF)和细胞间黏附分子-1(ICAM-1)蛋白表达的影响.方法 将HUVEC细胞分为正常对照组(NG)、高糖模型组(HG)及不同浓度黄诺玛苷组(50、100、200、400 μmol/L).酶联免疫吸附测定法(ELASA)检测细胞上清液中丙二醛(MDA)、超氧化物歧化酶(SOD)、过氧化氢酶(CAT)、谷胱甘肽过氧化物酶(GSH-Px)的活性.用Western blot方法检测细胞中VEGF和ICAM-1蛋白表达水平.结果 与NG组比较,HG组MDA明显升高,SOD、CAT、GSH-Px明显降低(P< 0.05或P< 0.01).与HG组比较,黄诺玛苷干预后,MDA含量显著降低(P<0.05),SOD、CAT、GSH-Px活力显著上升(P<0.05).Western blot结果显示,与NG组比较,HG组VEGF和ICAM-1蛋白表达量增多(P< 0.05或P<0.01).与HG组比较,黄诺玛苷干预后VEGF和ICAM-1蛋白表达量随浓度递增而依次减少(P< 0.05或P<0.01).结论 黄诺玛苷可能通过改善氧化应激水平和下调VEGF、ICAM-1的表达对高糖诱导HUVEC损伤具有一定保护作用.%Objective To observe the effects of flavanomarein glycosides of Coreopsis tinctoria Nutt.on oxidative stress and expression of vascular endothelial growth factor (VEGF) and intercellular cell adhesion molecule-1 (ICAM-1) protein in human umbilical vein endothelial cells (HUVEC) induced by high glucose.Methods The cultured HUVEC cells were divided into normal control group (NG group),high glucose model group (HG group) and different concentrations of flavanomarein group (50,100,200,400 μmol/L).The activity of malondialdehyde (MDA),superoxide dismutase (SOD),catalase (CAT) and glutathione peroxidase (GSH-Px) in supernatant were detected by enzyme-linked immunosorbent assay (ELASA).The expression levels of VEGF and ICAM-1 protein in cells were detected by Western blot method.Results Compared with NG group,the content of MDA in HG group was significantly increased,the levels of SOD,CAT and GSH-Px in HG group were significantly decreased (P < 0.05 or P < 0.01).Compared with HG group,after flavanomarein intervention,the content of MDA in HG group was significantly decreased,CAT,GSH-Px activity increased significantly (P < 0.05).The results of Western blot showed that compared with NG group,the expression of VEGF and ICAM-1 protein in HG group increased (P < 0.05 or P < 0.01).Compared with HG group,after flavanomarein intervention,the expression of VEGF and ICAM-1 protein increased with the increase of concentration followed by reduc tion (P < 0.05 or P < 0.01).Conclusion Flavanomarein has a protective effect on HUVEC induced by high glucose may be through improving the level of oxidative stress and decreasing the levels of VEGF and ICAM-1.
