重组减毒沙门氏菌Δcrp C79-13(pcDNA3-HN)在雏鸡体内主要组织器官定植情况检测

         

摘要

为检测重组减毒沙门氏菌Δcrp C79-13(pcDNA3-HN)在雏鸡体内主要组织器官定植情况,本研究将120只1日龄雏鸡随机分成3组,分别用减毒沙门氏菌Δcrp C79-13、重组减毒沙门氏菌Δcrp C79-13(pcDNA3-HN)和PBS 免疫,在免疫后4、24、48、72、96、120、144及168 h,分别检测减毒沙门氏菌Δcrp C79-13和重组减毒沙门氏菌Δcrp C79-13(pcDNA3-HN)在雏鸡心脏、肝脏、脾脏、盲肠和血液的定植情况。结果显示,在免疫后24 h 减毒沙门氏菌Δcrp C79-13在血液中数量达到了一个峰值,24~48 h 血液细菌数量下降,48~96 h 血液中细菌数上升并在96 h达到另一个峰值,96~168 h 血液中细菌数量下降,在心脏、肝脏、脾脏、盲肠这几个脏器中,细菌的数量分别在4~72 h呈上升趋势并在72 h 达到峰值,72~96 h 脏器细菌数量下降,96~120 h 菌数上升并在120 h 达到峰值,随后细菌数量下降;免疫重组减毒沙门氏菌Δcrp C79-13(pcDNA3-HN)组在雏鸡各脏器细菌总数变化及定植情况与Δcrp C79-13相似。结果表明,重组菌株Δcrp C79-13(pcDNA3-HN)与亲本菌株Δcrp C79-13均可在雏鸡心脏、肝脏、脾脏、盲肠等器官定植,且在各脏器中菌株定植变化趋势一致但数量下降。%The present study was designed to detect Δcrp C79-13 (pcDNA3-HN)which was a recombinational attenuated Salmonella embedded status in chicks major organs,120 one-day chicks were randomly assigned into 3 treatments according to a single factorial arrangement, which were immunized by Δcrp C79-13,recombinational Δcrp C79-13(pcDNA3-HN)and PBS,re-spectively.After immunization for 4,24,48,72,96,120,144 and 1 68 h,the embedded status ofΔcrp C79-13and recombinational Δcrp C79-13 (pcDNA3-HN )was detected in the heart,liver, spleen,cecum and blood of chicks.The results showed that the amount of attenuated immune Sal-monella Δcrp C79-13 reached a peak in the blood within 24 h,blood bacterial population decline in 24 to 48 h,number of bacteria in the blood rose at 48 to 96 h and reached another peak in 96 h, and the bacteria of the blood fell at 96 to 1 68 h .In the heart,liver,spleen and cecum,the number of bacteria in these organs at 4 to 72 h had an upward trend and reached a peak at 72 h,viscera bacterial population declined at 72 to 96 h,and the count rose at 96 to 120 h and reached a peak at 120 h,then the number of bacteria decreased.It was similar to the change in each viscera strain of group Δcrp C79-13(pcDNA3-HN).These data indicated that both of the recombinational bacterial strains Δcrp C79-13(pcDNA3-HN)and parental bacterial strainΔcrp C79-13 could be embedded in a series of organ of chicks such as heart,liver,spleen,and cecum.Meanwhile,there was a consist-ent declining trend of bacterial strain amount in each viscus of chicks.
