大孔树脂分离纯化隐丹参酮的研究

         

摘要

To optimize the optimum conditions for the isolation and purification of cryptotanshinone from tanshinone extract, using cryptotanshinone content detected by HPLC as index, the kind of macroporous resins, adsorption conditions and elution conditions were investigated.The optimized conditions were as follows:LSD-ro type macroporous resin ,pH value 3, time of duration 3h, with gradient elutin,first 60 % ethanol elution about 50 mL ,then 80%ethanol elution, elution volume of 190mL.The concentration of tanshinone from theextract can be increased from 21 % to more than 45% under these conditions, and the concentration was increased about 1.14 times.%为了优选大孔树脂纯化丹参药材丹参酮提取液中隐丹参酮的工艺,以高效液相色谱检测出的隐丹参酮含量为指标,对大孔树脂型号、吸附条件、洗脱条件进行了考察.优选出的最佳条件为选取LSD-40型大孔树脂,上样液pH值3,上样液静置时间3 h,采用梯度洗脱,先用60%乙醇洗脱50mL左右,再用80%乙醇溶液洗脱,洗脱液体积为190 mL左右,可以将隐丹参酮的质量浓度由提取液中的21%左右富集到45%以上,提高了约1.14倍.
