薄层等电聚焦-X-射线荧光光谱法测定铁超载小鼠肝中铁含量分布

     

摘要

The distribution of iron content in iron overload mouse liver was studied by using thin-layer isoe-lectric focusing and X-ray fluorescence spectrometry. The result showed that the iron content in iron overload mouse liver cytosol, mitochondrion and microsome significantly increased,and the iron content increasing order in the subcellular fractions was microsome>cytosol> mitochondrion. Meanwhile, the results also showed that the pi values of iron-containing proteins in cytosol were 3. 0,4. 7 and 5. 7,while in mitochondrion were 5. 7 and 7. 0,and in microsome were 3. 0,5. 7 and 7. 3. The contents of all iron-containing protein were significantly increased in cytosol and mitochondrion,but the pI value of increased iron-containing protein in microsome was 5.7.%采用薄层等电聚焦-X-射线荧光光谱法对铁超载小鼠肝中铁含量的分布进行了研究.结果表明:机体摄入过量铁,肝胞液、线粒体以及微粒体中铁含量都较正常小鼠显著增加,且增加的量存在如下关系:微粒体>肝胞液>线粒体;肝胞液中含铁蛋白主要分布在等电点3.0、4.7和5.7处,线粒体中含铁蛋白主要分布在等电点5.7和7.0处,微粒体含铁蛋白主要分布在等电点3.0、5.7和7.3处;铁超载后,肝胞液和线粒体各处铁含量均显著增加,而微粒体中所增加的铁主要集中在等电点5.7处.
