薰衣草精油提取残渣中多酚的提取纯化工艺研究

     

摘要

Using total phenolic content(TPC)as index,the extraction conditions of lavender polyphenols from extraction residue of lavender essential oil were optimized based on following factors:extraction solvent, solid-liquid ratio,extraction time and extraction temperature.Then the purification of lavender polyphenols was studied by using AB-8 macroporous resin as separation material.The results showed that the best solvent was 70% acidified acetone [V(acetone)∶V (HCl,1 mol·L-1 )=70∶30].The extraction yield of polyphenols was the highest when the solid-liquid ratio was 1∶20(g∶mL),extraction time was 1 h and extraction temperature was 40 ℃.The adsorption kinetic results showed that,the saturable adsorption of AB-8 macroporous resin was 2.5 mg·g-1 ,the adsorption rate reached more than 90% within 10 min and reached saturation in 20 min by controling the mass ratio of AB-8 resin and total phenolics at 450∶1,desorption could be completed with in 10 min with the desorption rate of 100% when methanol aqueous solution was used as elution solvent.And the lav-ender polyphenols could be completely eluted by 60% methanol solution.%以总酚含量为考察指标,从提取溶剂、料液比、提取时间和提取温度4个方面对薰衣草精油提炼残渣中的多酚提取工艺进行了优化,并对 AB-8大孔树脂对薰衣草多酚的纯化工艺进行了研究。结果表明:以70%丙酮酸[V (丙酮)∶V (HCl,1 mol·L-1)=7∶3]作提取溶剂,在料液比为1∶20(g∶mL)、40℃下水浴加热1.0 h 时多酚提取率最高;吸附动力学结果表明,按照 AB-8大孔树脂与薰衣草多酚的质量比为450∶1上样时,在10 min 内吸附率达到90%以上,20 min 左右可达到吸附饱和,饱和吸附量约为2.5 mg·g-1;解附实验表明,用甲醇和水的混合溶液进行梯度洗脱时,10 min 内即可达到100%的解附率,其中60%的甲醇水溶液可将薰衣草多酚完全洗脱出来。
