Using the activity of antibacterial substance of the fermentation broth from marine Actinomycete AM8 as an index,a single factor experiment was used to optimize the basic fermentation culture medium and fermentation conditions.The antibacterial active substance of the fermentation broth was preliminarily studied.The optimal culture medium was as follows:tryptone 17 g·L-1,soybean peptone 3 g·L-1,sucrose 2.5 g·L-1,NaCl 5 g·L-1,K2HPO4 2.5 g·L-1,and sea crystal 20 g·L-1.The optimal fermentation conditions were as follows:pH value 7,temperature 28 ℃,inoculum 5%,shaking speed 200 r·min-1,and fermentation time 5 d.The antibacterial active substance was medium polarity substance,which had good acid resistance and certain thermal stability,and easily became inactivated under strong alkalis and high temperature conditions.%以海洋放线菌AM8发酵液抑菌物质的活性为评价指标,采用单因素实验对海洋放线菌AM8的基础发酵培养基和发酵条件进行优化,并对抑菌活性物质进行了初步研究.确定最佳培养基为:胰蛋白胨17 g·L-1,大豆胨3 g·L-1,蔗糖2.5 g·L-1,NaCl 5 g·L-1,K2HPO4 2.5 g·L-1,海盐20 g·L-1;最佳发酵条件为:pH值7,温度28 ℃,接种量5%,转速200 r·min-1,发酵时间5 d.该抑菌活性物质为中等极性物质,具有良好的耐酸性和一定的热稳定性,在强碱、高温环境下易失活.
